摘要
目的探讨清热明目颗粒对实验性自身免疫性葡萄膜炎(EAU)大鼠的治疗作用以及对炎症细胞因子表达的影响。方法24只Lewis大鼠随机分为空白组、EAU组、实验组,雷公藤组4组,每组各6只。利用光感受器间维生素A结合蛋白(IRBP)免疫大鼠制作EAU模型,空白组注射等量不含IRBP的乳化液。于免疫的同时,实验组给予3.5 g/(kg·d)清热明目颗粒悬浊液灌胃;雷公藤组给予6.8μg/(kg·d)含雷公藤甲素的悬浊液灌胃;空白组和EAU组给予同等剂量蒸馏水灌胃。各组给药均为每日1次,连续7 d。各组大鼠在炎症高峰期(第15 d)摘取眼球制备视网膜切片,根据Caspi组织学分级法评分,分离各组大鼠脾脏和腹部淋巴结淋巴细胞,采用ELISA法检测IL-2、IL-10、IL-17、IFN-γ表达水平。结果(1)炎症评分:EAU组、实验组和雷公藤组均于免疫后8 d开始发病,EAU组炎症反应评分明显增高,与空白组差异有统计学意义(t=49.019,P=0.000),而实验组和雷公藤组与空白组差异无统计学意义。免疫第10 d,各组炎症反应均升高,与空白组差异均有统计学意义(tEAU组=7.904,P=0.000,t实验组=3.980,P=0.001,t雷公藤组=7.250,P=0.000),而实验组评分低于EAU组(t=3.925,P=0.001)和雷公藤组(t=3.271,P=0.004),差异有统计学意义。免疫第15 d,各组炎症反应达高峰,实验组评分低于EAU组(t=7.071,P=0.000),差异有统计学意义。(2)组织病理学:实验组视网膜有炎症细胞浸润和结构破坏较轻,组织病理学评分低于EAU组(t=10.675,P=0.000),和雷公藤组(t=5.115,P=0.000),差异有统计学意义。(3)炎性细胞因子:EAU组淋巴细胞培养上清液IL-2、IL-17、IFN-γ表达升高,IL-10表达降低(FIL-2=912.986,F IL-17=103.286,FINF-γ=1852.132,FIL-10=291.765,均P=0.000),均有统计学意义;与EAU组比较,淋巴细胞培养上清液IL-2、IL-17、IFN-γ表达量在实验组(t_(IL-2)=31.041,t_(IL-17)=7.369,t_(INF-γ)=16.363,均P=0.000)和雷公藤组(t_(IL-2)=17.137,tIL-17=8.239,tINF
OBJECTIVE To investigate the therapeutic effect of Qingremingmu Granule on experimental autoimmune uveitis(EAU)rats and the expression of key inflammatory cytokines.METHODS Twenty-four Lewis rats were randomly divided into four groups:blank group,EAU group(EAU),experimental group(EXP)and Tripterygium wilfordii group(TW),with six rats in each group.The EAU model was established by immunizing rats with interphotoreceptor retinoid-binging protein(IRBP).The blank group was injected with the same amount of emulsion without IRBP.While immunization,the EXP group was given 3.5 g/(kg·d)Qingremingmu Granule suspension by intragastric administration;the TW group was given 6.8μg/(kg·d)suspension containing triptolide by intragastric administration;The blank group and the EAU group were given the same dose of distilled water by intragastric administration.Each group was administered once per day for 7 consecutive days.The eyeballs of rats in each group were extracted for the preparation of retinal sections at the peak of inflammation(the 15th day),which were scored according to Caspi histological grading method.Lymphocytes from spleen and abdominal lymph nodes of each group were separated,and the expression levels of IL-2,IL-10,IL-17 and IFN-γwere detected by ELISA.RESULTS(1)Inflammatory scores:The EAU group,the EXP group and the TW group all began to develop symptoms on 8th day after immunization.The inflammatory response scores of the EAU group were significantly higher than those of the blank group(t=49.019,P=0.000),while there was no significant difference between the EXP group and the TW group when compared to the blank group.On the 10th day of immunization,the scores of inflammatory response increased in all groups,with statistically significant differences compared with the blank group(tEAU=7.904,P=0.000,tEXP=3.980,P=0.001,tTW=7.250,P=0.000).The scores of the EXP group were lower than those of the EAU group(t=3.925,P=0.001)and the TW group(t=3.271,P=0.004),and the differences were statistically significant.On the 1
