摘要
目的探讨不同特殊染色方法对关节软骨组织形态显示的效果比较。方法选取肩袖修复术后8周的兔8只,截取双肩关节,行纵切面切开后固定、脱钙、石蜡包埋,切片,分别行改良Harris′苏木精-伊红染色、番红O固绿染色、苦味酸天狼星红染色、改良Masson染色、爱尔新兰(pH 25)-高碘酸-无色品红法(AB-PAS)染色,观察兔肩关节软骨组织形态学变化并进行方法学比较。结果改良Harris′苏木精-伊红染色缺乏组织特异性,并不能清晰反映软骨的分层结构,可作初筛染色来选择;番红O固绿染色操作简便、颜色鲜艳、对比清晰,能全面反映软骨组织学信息;苦味酸天狼星红染色结构分层、颜色对比不明显,且需特殊设备,实用性和经济性不高;改良Masson染色操作步骤繁琐,费时,软骨在蓝染的情况下,软骨层的结构分层并不算十分明显,对比染色略逊于番红O固绿染色;AB-PAS染色操作繁琐,染色结果只能部分反映关节软骨的多糖蛋白分布,并未能显示软骨生长的全面信息。结论番红O固绿染色可以获得更为全面的关节软骨形态学信息,对比清晰。
Objective To investigate the effect of different special staining methods on the display of articular cartilage morphology.Methods Eight rabbits were selected at 8 weeks after rotator cuff repair.The shoulder joints were amputated,fixed after lengthwise cut,decalcified and embedded in paraffin,sectioned.Modified Harris hema-toxylin-eosin(HE)staining,safranin O-fast green staining,picrosirius red staining,modified Masson staining,and AB-PAS staining were performed,respectively.The histomorphological changes in the cartilage of shoulder joint were de-termined and the methods were compared.Results Modified Harris HE staining lacked tissue specificity and could not clearly reflect the layered structure of cartilage,which could be selected as a primary screening staining.Safranin O-fast green staining had the advantages of simple operation,bright color and clear contrast,which could fully reflect the histological information of cartilage.Picrosirius red staining had no obvious structure stratification and color con-trast,and required special equipment,which was not practical and economical.The operation of modified Masson stain-ing was cumbersome and time-consuming.In the case of blue staining of cartilage,the structural stratification of the cartilage layer was not very obvious,and the contrast staining was slightly inferior to safranin O-fast green staining.The operation of AB-PAS staining is cumbersome,and the staining results could only partially reflect the polyglyco-protein distribution of articular cartilage,and failed to show the comprehensive information of cartilage growth.Conclusion Safranin O-fast green staining may obtain more comprehensive morphological information of articular cartilage with clear contrast.
