摘要
目的:观察黄芪多糖对食管鳞状细胞癌细胞肌动蛋白细胞骨架的影响。方法:利用荧光素异硫氰酸酯标记的鬼笔环肽标记食管鳞状细胞癌KYSE-150的纤丝状肌动蛋白(F-actin),经激光共聚焦显微镜观测黄芪多糖诱导后KYSE-150细胞内F-actin骨架的变化。结果:KYSE-150的细胞膜F-actin骨架呈粗大束状。经黄芪多糖诱导后,细胞膜内层F-actin骨架变得纤细、丰富,大量的F-actin纤维支撑起细胞膜,促使细胞膜形成丰富的微绒毛状结构,增加了细胞膜间的接触面积,细胞间不易相互侵入。结论:黄芪多糖诱导食管鳞状细胞癌KYSE-150细胞F-actin骨架的重构,有可能促使癌细胞的良性转化。
Objective:To investigate the effect of astragalus polysaccharide on the actin cytoskeleton of esophageal squamous cell carcinoma cells.Methods:F-actin of esophageal squamous cell carcinoma KYSE-150 was labeled with Phalloidin-FITC,and the changes of F-actin cytoskeleton in KYSE-150 cells after astragalus polysaccharide induction were observed by laser confocal microscopy.Results:The cell membrane F-actin skeleton of KYSE-150 was in the form of thick bundles.After induced by astragalus polysaccharide,the inner layer of cell membrane F-actin skeleton became slender and abundant,and a large number of F-actin fibers supported the cell membrane,prompting the formation of abundant microvilli-like structures,which increased the contact area between cell membranes and cells were not easily invaded by each other.Conclusion:Astragalus polysaccharide induces F-actin skeleton remodeling in esophageal squamous cell carcinoma KYSE-150 cells,which may promote the benign transformation of cancer cells.
