摘要
目的筛选心脏外科手术后房颤(POAF)的差异表达基因,并进行功能分析。方法从GEO数据库获取1个基因表达谱数据集GSE143924,样本来源于心外科手术患者心外膜脂肪组织。根据术前特征对15例POAF患者(病例组)和15例术后窦性心律患者(对照组)进行匹配。通过GEO2R在线分析病例组和对照组差异表达基因,对差异表达基因进行GO分析及KEGG分析,构建蛋白与蛋白相互作用(PPI)网络,筛选Degree前10位的基因为POAF的关键基因。结果共筛选获得病例组和对照组的254个差异表达基因,病例组较对照组表达上调82个、表达下调172个。GO和KEGG分析显示,差异表达基因涉及的信号通路主要与补体和凝血级联等相关,生物学过程主要集中在细胞黏附、免疫反应等。PPI网络筛选得到10个关键基因分别为TLR2、CDH2、VTN、CCL2、KDR、C1QA、BMP2、ZAP70、HP、CFB,这些基因主要调控炎症反应及免疫反应。结论POAF差异表达基因涉及的信号通路主要与补体和凝血级联等相关,生物学过程主要集中在细胞黏附、免疫反应等,炎症反应和免疫反应在POAF的发生发展中起关键作用。
Objective To screen differentially expressed genes(DEGs)of postoperative atrial fibrillation(POAF)after cardiac surgery and to analyze their functions.Methods A gene expression profile data set GSE143924 was obtained from GEO database.The sample was from epicardial adipose tissues of patients undergoing cardiac surgery.According to the preoperative features,15 patients with POAF(case group)and 15 patients with sinus rhythm after operation(control group)were matched.Then we analyzed the differential expressed genes between the case group and the control group by GEO2R online.Moreover,GO and KEGG enrichment analysis of DEGs were conducted,and the top 10 genes of Degree were selected as the key genes of POAF.Results A total of 254 differentially expressed genes were screened out from the case group and control group,of which 82 were up-regulated and 172 were down-regulated.GO and KEGG analysis showed that the signal pathways involved in DEGs were mainly related to complement and coagulation cascade,and the biological processes principally focused on cell adhesion and immune response.Ten key genes screened out by PPI network were TLR2,CDH2,VTN,CCL2,KDR,C1QA,BMP2,ZAP70,HP and CFB,which mainly regulated the inflammatory and immune responses.Conclusions The signal pathways involved in POAF DEGs are mainly related to complement and coagulation cascade,and the biological processes mainly focus on cell adhesion and immune response.Inflammation and immune response play a key role in the development and progression of POAF.
