摘要
本研究旨在筛选出1株纤维素降解真菌并探究其产酶活性,为西藏纤维素的降解提供菌种资源,也为下一步复合菌群的构建奠定基础。选用刚果红染色法进行初筛,采用滤纸崩解试验和酶活力测定试验完成复筛。结果表明,在8个供试菌株中,菌株LZ2007-1,其内切酶活力、外切酶活力、β-葡聚糖苷酶活力和滤纸酶活力峰值分别为54.14 U/mL、53.67 U/mL、30.37 U/mL和42.99 U/mL。结果说明,该菌株具有进一步应用于生产的价值,本试验为该菌株的深入开发利用提供了理论依据,也丰富了纤维素生物降解的菌种库。
The purpose of this research is to screen out a cellulose-degrading fungus and explore its enzyme-producing activity, so as to provide bacterial resources for the degradation of cellulose in Tibet, and to form a foundation for the next step of the construction of the complex flora. In specific, the Congo red staining method was used for the primary screening, and the filter paper disintegration test and enzyme activity determination test were used to complete the re-screening.The results showed that among the 8 tested strains, the strain LZ2007-1 performed best, where the endonuclease activity, exonuclease activity, β-glucosidase activity and filter paper enzyme activity peak values reached 54.14 U/mL, 53.67 U/mL, 30.37 U/mL and 42.99 U/mL, respectively. These results indicated that this strain had the value of further application in actual production. This experiment could provide a theoretical basis for the in-depth development and utilization of this strain, and also enriche the strain library for cellulose biodegradation.
