摘要
目的探讨三叶因子3(trefoil factor 3,TFF3)在小鼠颌下腺生后发育中的表达及作用。方法运用HE染色、PAS染色及免疫组织化学法对生后1~31 d的仔鼠颌下腺组织切片进行染色及形态学观察;Western blot检测TFF3变化趋势。结果①HE染色生后1 d小鼠颌下腺可观察到细胞核分裂象;7 d腺泡和导管染色出现区别,腺泡细胞胞浆嗜碱性较强;15 d可区分浆液性和黏液性腺泡;②腺泡细胞PAS染色阳性,1~15 d强度随日龄增加而增强,导管比例增加,15 d后PAS阳性信号基本类似;③免疫组化染色TFF3阳性信号位于导管,腺泡呈阴性;1~5 d导管上皮细胞胞浆呈TFF3弱阳性表达,7~31 d导管阳性信号逐渐增强,15 d可在管腔内观察到TFF3阳性分泌物;④Western blot3~5 d颌下腺组织已存在低水平TFF3表达,7~31 d TFF3表达随日龄增加不断增强。结论小鼠颌下腺在生后15 d以后逐渐发育完善,TFF3阳性信号表达于导管上皮,15 d后导管腔内出现TFF3,可能具有促进导管细胞发育和保护口腔黏膜的作用。
Objective To study the expression and role of trefoil factor 3(TFF3)in postnatal development of mice submandibular gland.Methods Tissue sections of mice submandibular gland from postnatal(P)1 d to 31 d were used to observe morphological changes by HE,PAS staining and immunohistochemistry methods.Expression trend of TFF3 was detected by Western blot.Results①HE staining:Under light microscope after HE staining,cell karyomitosis was observed on the P1 d in submandibular gland of mice.The staining of acinar and duct was different on the P7 d,and the basophilic cytoplasm of acinar cells was stronger;serous and mucinous acinars could be distinguished on the P15 d.②PAS staining:PAS staining of acinar cells was positive,and staining intensity and proportion of ducts continuously increased with age from P1 d to P15 d,and PAS positive signal was similar after P15 d.③Immunohistochemistry staining:TFF3 positive signal was located in the duct and the acinus was negative.TFF3 signal was weakly positive from P1 d to P5 d in cytoplasm of ductal cells,TFF3 positive signal in ducts gradually increased from P7 d to P31 d,and TFF3 positive secretion could be observed in the lumen from P15 d.④Western blot:The expression of TFF3 peptide was low in submandibular gland from P3 d to P5 d,and increased with age from P7 d to P31 d.Conclusion Submandibular gland developed gradually after P15 d,and TFF3 positive signal mainly expressed in ductal epithelial cells and TFF3 appeared in the lumen after P15 d.TFF3 may have the roles of promoting ductal cells growth and protecing the oral mucosa.
