摘要
为加快‘由良’蜜橘的脱毒效率,以‘卡里佐’枳橙为试管砧木,探究了试管播种培养砧木的处理方法、成年态茎段消毒方法、单芽茎段培养方法和不同茎尖来源利用率,以期找到最优的方式来获取数量多、质量好的茎尖。结果表明:划破内种皮砧木种子污染率为32.5%,萌芽率为60.0%,获得的无菌试管砧木最多;培养室培养植株后再进行成年态茎段培养的污染率显著低于从田间采样,从培养室取接穗,使用1%NaClO消毒20min后污染率为20.0%,褐化率为10.0%,萌芽率为70.0%;MS+1.0 mg/L 6-BA+0.05 mg/L IBA的培养基对‘由良’单芽茎段离体培养效果最好,萌芽率为91.6%,增殖倍数为3.0;但使用MS培养基培养多芽茎段获得茎尖的方式,其不定芽的利用率最高,为81.8%。
In order to speed up the virus-free efficiency of’Youliang’tangerine,the’Carrizo’aurantii was used as the test tube rootstock and the treatment method of in vitro seeding rootstock was studied,disinfection methods of adult stem segments,cultivation methods of single-bud stem segments and utilization rates of different stem tip sources,in order to find the optimal experimental method to obtain large quantity and good quality stem tips.The results showed that the seed contamination rate and germination rate of rootstocks were 32.5%and 60.0%respectively,the most sterile test tube rootstocks were obtained.The contamination rate of adult stem segment culture after plant culture in culture room was significantly lower than that of field sampling.The contamination rate,browning rate and germination rate of scion were 20.0%,10.0%and 70.0%after 1%Na Cl O disinfection for 20 min.The culture medium MS+1.0 mg/L 6-BA+0.05 mg/L IBA had the best effect on the single bud segment of’Youliang’in vitro.The germination rate was 91.6%and the proliferation rate was 3.0.However,the utilization rate of MS culture medium to obtain shoot tips was the highest,which was 81.8%.
