基于形态指标和转录组的小麦抗旱与耐盐相关性分析 被引量:2

摘要 干旱和盐害是影响小麦产量的两大非生物逆境,明确小麦抗旱、耐盐性的相关关系对抗旱耐盐品种的培育具有重要意义。研究以173份小麦品种(系)为材料,利用20%的PEG和150 mmol/L的NaCl溶液分别模拟干旱和盐胁迫,在芽苗期测定相对胚芽鞘长、相对根长、相对苗高、相对发芽率、相对鲜质量等形态指标,采用隶属函数法对其进行抗旱、耐盐性综合评价,并对抗旱与耐盐的相关性进行分析。结果表明,小麦抗旱性与耐盐性的相关系数为0.43,呈中度正相关关系。利用转录组测序技术对抗旱耐盐品种青麦6号干旱、盐胁迫48 h叶片分析,结果表明,干旱、盐胁迫共上调基因数分别占干旱、盐胁迫上调基因数的35.48%和59.22%,干旱、盐胁迫共下调基因数分别占干旱、盐胁迫下调基因数的35.58%和88.46%;GO功能富集结果显示,干旱、盐胁迫上调基因显著富集前20个亚类相同的有18个,干旱、盐胁迫下调基因显著富集前20个亚类相同的有19个。说明小麦抗旱性与耐盐性的遗传调控机制有一定的相似性。 Drought and salt damage are two abiotic stresses that affect wheat yield.It is of great significance to clarify the relationship between wheat drought resistance and salt tolerance for the breeding of drought-resistant and salt-tolerant cultivars.In this study,173 wheat cultivars(lines)were used as materials,and 20%PEG and 150 mmol/L NaCl solutions were applied to simulate drought and salt stress,respectively.The morphological indexes,such as relative coleoptile length,relative root length,relative seedling height,relative germination rate and relative fresh weight,were measured at the bud and seedling stage.Then the abilities of drought resistance and salt tolerance were comprehensively evaluated by membership function method.Lastly,the correlation analysis between them was calculated.The result showed that the correlation coefficient was 0.43,indicating a moderate positive correlation.Besides,the leaves of Qingmai 6 seedlings treated with drought or salt for 48 h were used for transcriptome analysis.The results showed that the co-upregulated genes accounted for 35.48%and 59.22%of up-regulated genes under drought and salt stress,respectively,while the co-downregulated genes accounted for 35.58%and 88.46%of down-regulated genes under drought and salt stress,respectively.GO enrichment analysis showed that the intersection number of the first 20 functional subclasses enriched by up-regulated genes under drought and salt stresses was 18,while it was 19 for the down-regulated genes.All these indicated that there is some similarity in the genetic regulation mechanism between drought resistance and salt tolerance in wheat.
