川芎提取物对血管紧张素Ⅱ诱导的肾炎症大鼠的影响 被引量:5

Effect of Chuanxiong extract on renal inflammation induced by angiotensinⅡ

摘要 目的探究川芎提取物对血管紧张素Ⅱ(AngⅡ)诱导的炎症反应及对微小RNA-103a-3p(miR-103a-3p)的影响。方法将健康雄性Wistar大鼠随机分为对照组、模型组和低、中、高剂量实验组。模型组16只,进行肾炎症模型造模处理。低、中、高剂量实验组各16只,在模型组的基础上给予10,20,40μmol·L^(-1)川芎提取物28 d,灌胃,另取16只健康雄性Wistar大鼠作为对照组,用等量的生理盐水连续灌胃28 d。各组每周称重1次,留取24 h尿液,于第28天处死各组大鼠,大鼠处死后取左肾标本。用Tail-cuff体积描记法的测定尾动脉收缩压,用全自动生化分析仪测定血肌酸酐(SCr)、血清白蛋白(ALB)、尿素氮(BUN)浓度,用酶联免疫吸附(ELISA)法和蛋白质印迹(Western Blot)法检测各组大鼠组织中炎症因子白细胞介素-1β(IL-1β)、肿瘤坏死因子-α(TNF-α)和白细胞介素-6(IL-6)水平,用实时荧光定量逆转录聚合酶链反应(qRT-PCR)法检测各组大鼠组织中miR-103a-3p的相对表达水平。结果对照组、模型组和低、中、高剂量实验组血肌酸酐分别为(18.76±2.31),(40.77±5.18),(31.66±2.64),(27.35±2.36),(23.31±2.87)μmol·L^(-1),血清白蛋白分别为(31.70±1.15),(20.06±1.25),(23.19±1.27),(26.98±1.33),(29.74±1.09)g·L^(-1),尿素氮分别为(4.76±0.63),(9.14±1.02),(7.53±0.84),(6.96±0.47),(5.01±0.51)mmol·L^(-1);IL-1β水平分别为(7.25±0.68),(37.46±3.51),(26.98±1.45),(20.19±2.03),(14.22±1.56)ng·mL^(-1),TNF-α水平分别为(0.82±0.09),(9.12±0.92),(6.19±0.51),(4.35±0.36),(2.19±0.39)ng·mL^(-1),IL-6水平分别为(124.32±10.34),(187.24±17.65),(160.29±13.67),(145.23±15.36),(132.18±11.23)ng·mL^(-1);miR-103a-3p水平分别为1.00±0.12,2.39±0.21,2.06±0.14,1.82±0.17,1.58±0.13。模型组大鼠在第1、2、3、4周血压明显高于对照组,低、中、高剂量实验组大鼠在第1、2、3、4周血压明显低于模型组(均P<0.05)。与对照组相比,模型组大鼠血肌酸酐、尿素氮、IL-1β� Objective To explore the effect of Chuanxiong extract on the inflammatory response induced by angiotensinⅡ(AngⅡ)and on microRNA-103a-3p(miR-103a-3p).Methods Healthy male Wistar rats were randomly divided into control group,model group and low,medium and high dose experimental groups.Sixteen rats in model group were established nephritis model,16 rats each in low,medium,high dose experimental group were treated with 10,20,40μmol·L^(-1) Chuanxiong extract by gavage on the basis of model group for 28 days.The other 16 healthy male Wistar rats were taken as control group and given equal amount of normal saline by gavage for 28 days.Each group was weighed once a week and urine was retained for 24 h.The rats in each group were sacrificed on day 28,and the left kidney specimens were taken after the rats were sacrificed.The systolic blood pressure of the tail artery was measured by Tail-cuff volumetric tracing method,and the concentrations of serum creatinine(SCr),serum albumin(ALB)and blood urea nitrogen(BUN)were measured by fully automatic biochemical analyzer,and the levels of inflammatory factors in the tissues of each group of rats were detected by enzyme-linked immunosorbent assay(ELISA)and Western Blot.The levels of interleukin-1β(IL-1β),tumor necrosis factor-α(TNF-α)and interleukin-6(IL-6)were measured by ELISA and Western Blot.Real-time fluorescent quantitative polymerase chain reaction(qRT-PCR)were used to detect the relative expression levels of miR-103a-3p in rat tissues in each group.Results The sereum creatinine of control group,model group and low,medium and high dose experimental groups were(18.76±2.31),(40.77±5.18),(31.66±2.64),(27.35±2.36),(23.31±2.87)μmol·L^(-1),ALB were(31.70±1.15),(20.06±1.25),(23.19±1.27),(26.98±1.33),(29.74±1.09)g·L^(-1),BUN were(4.76±0.63),(9.14±1.02)mmol·L^(-1),(7.53±0.84),(6.96±0.47),(5.01±0.51)mmol·L^(-1);IL-1βlevels were(7.25±0.68),(37.46±3.51),(26.98±1.45),(20.19±2.03),(14.22±1.56)ng·mL^(-1),TNF-αlevels were(0.82±0.09),(9.12±0.92),(6.19
