摘要
目的:比较柱切换-HPLC和均相酶免法快速测定艾司西酞普兰血药浓度结果的准确性。方法:建立柱切换HPLC法和均相酶免法测定艾司西酞普兰的血药浓度的方法,并对两种方法进行方法学验证。收集本院住院患者60例病例,两种方法同时监测同一批患者的艾司西酞普兰血药浓度。结果:柱切换-高效液相色谱法与均相酶免法进行线性回归,回归方程为:y=0.9219x+0.0501(n=60,r=0.8999)。两种方法比较的结果进行T检验,同一患者测定结果无显著性差异(p>0.05)。结论:柱切换HPLC和均相酶免法均可测定艾司西酞普兰血药浓度,但是均相酶免法测定复杂,成本高,柱切换HPLC法具有灵敏度高、简单、方便、且显著降低检测成本等优点,可适用于医院检验科和临床药学的应用。
Objective:To compare the accuracy of column switching HPLC and homogeneous enzyme immunoassay for the rapid determination of Escitalopram in blood.Methods:Column switching HPLC and homogeneous enzyme immunoassay were established to determine the plasma concentration of escitalopram,and the two methods were verified.60 inpatients in our hospital were collected.The plasma concentration of escitalopram in the same batch of patients was monitored by two methods at the same time.Results:The linear regression between column switching HPLC and homogeneous enzyme immunoassay was y=0.9219 x+0.0501(n=60,r=0.8999).The results of the two methods were compared by t-test,and there was no significant difference in the results of the same patient(p>0.05).Conclusion both column switching HPLC and homogeneous enzyme immunoassay can determine the plasma concentration of escitalopram,but the homogeneous enzyme immunoassay is complex and costly.On the contrary,column switching HPLC has the advantages of high sensitivity,simplicity,convenience and significantly reducing the detection cost.It can be used in hospital laboratory and clinical pharmacy.
