小鼠耳芥CONSTANS-like基因家族的鉴定及响应干旱胁迫表达分析

摘要 CONSTANS-like家族蛋白是植物光周期途径的转录因子,在植物的生长发育和胁迫应答等过程中起着重要的作用。本研究利用HMMER3.0在小鼠耳芥(Arabidopsis pumila)全基因组范围内进行ApCOL基因家族成员分析,共得到34个ApCOL家族成员。系统进化分为3个亚组,每组ApCOL蛋白质均具有典型的B-box和CCT结构域。亚细胞定位预测结果显示ApCOL成员均定位在细胞核中。基因结构分析表明,ApCOL的外显子、内含子结构和长度在各亚组内具有同一性。26个ApCOL基因与拟南芥(Arabidopsis thaliana)存在共线性关系。ApCOL基因上游启动子序列存在多种光响应、环境胁迫、生长发育以及激素应答等相关顺式作用元件;通过qRT-PCR分析,发现12个ApCOL表达量在PEG6000胁迫下显著上调,暗示这些ApCOL基因参与干旱胁迫响应。本研究丰富了ApCOL家族转录因子的信息,为更好的研究小鼠耳芥生长发育和胁迫响应机制提供一定的参考。 CONSTANS-like family proteins are transcription factors of plant photoperiod pathways,which play important roles in plant development and stress response.In this study,The CONSTANS-like gene family in Arabidopsis pumila were characterized in genome-wide using HMMER3.0 program.A total of 34 ApCOL family members were identified.Phylogenetic analysis revealed that these genes were divided into three subgroups,and all ApCOL proteins contained B-box and CCT domains.The subcellular localization prediction showed that ApCOL proteins were located in the nucleus.Gene structure analysis showed that the structure and length of the exons and introns of ApCOL s were similar in one subgroup.The 26 ApCOL genes had collinearity relationships with Arabidopsis thaliana.There are various cis-acting elements in the upstream promoter sequence of ApCOL genes,such as light response,environmental stress,growth and development,and hormone response.Gene transcription analyses using qRT-PCR revealed that the expression levels of 12 ApCOL genes were significantly up-regulated in response to PEG6000 stress,suggesting that these ApCOL genes were involved in drought stress response.This study enriched the information of ApCOL family transcription factors,and provided references for better understanding of the growth and development and stress response mechanisms in ephemeral plant Arabidopsis pumila.
