摘要
过氧化亚硝酰阴离子(ONOO^(-))是由一氧化氮(NO)和超氧化物自由基(O_(2)^(·-))在扩散控制下自由组合产生的,它是一种强氧化剂和硝化剂,会对细胞中的DNA、蛋白质和其他生物分子造成损害。由于ONOO^(-)在生理条件下寿命短(≈10 ms)、反应活性高、浓度低、在生物系统中发挥的确切致病作用尚未完全清楚,因此开发对ONOO^(-)灵敏度高、选择性好、实时的检测方法具有十分重要的意义。本文主要综述了近年来在开发检测ONOO^(-)的小分子荧光探针方面取得的进展,以与ONOO^(-)反应的机制分类,分别简述了基于硫族元素化合物氧化反应、碳碳双键断裂反应、酰肼的氧化反应、氧化N⁃脱芳基化反应、硼酸或硼酸酯的氧化反应等反应机制的荧光探针。
Peroxynitrite(ONOO^(-))is produced by free combination of nitric oxide(NO)and superoxide radicals(O_(2)^(·-))under the diffusion control.It is a strong oxidant and highly effective nitrifying agent,which can damage the DNA,proteins and other biological molecules in cells.Peroxynitrite has short lifespan(≈10 ms),high reactivity,and low concentration under physiological conditions,while its exact pathogenic role in biological systems has not been fully elucidated.Therefore,it is of great significance to develop a detection method with high sensitivity,high specificity and real⁃time for ONOO^(-).This article mainly reviewed the progress made in the development and detection of ONOO^(-)fluorescent probes in recent years.Classified by mechanism of reaction with ONOO^(-),the fluorescent probes were presented based on the oxidation of chalcogenides,cleavage of C=C double bonds,oxidation of hydrazides,oxidative N⁃dearylation,oxidation of boronic acids or boronates and other reaction strategies.
