摘要
目的建立复方黄芪乳膏HPLC指纹图谱,并同时测定3种成分含量。方法对复方黄芪乳膏75%甲醇提取液进行分析,采用Agilent 5 TC-C_(18)(4.6 mm×250 mm,5μm)色谱柱,流动相为甲醇-0.1%甲酸水溶液,梯度洗脱,检测波长250 nm,柱温30℃,体积流量0.8 mL·min^(-1)。结果建立了复方黄芪乳膏HPLC指纹图谱,标定了18个共有峰,共鉴定出京平尼苷酸、咖啡酸、羟基红花黄色素A、毛蕊异黄酮葡萄糖苷、毛蕊花糖苷及肉桂酸6个色谱峰,10批样品的相似度均大于0.930。京尼平苷酸、毛蕊异黄酮葡萄糖苷、肉桂酸的含量分别为1.59~1.84 mg·g^(-1)、1.37~1.58 mg·g^(-1)、0.791~0.943 mg·g^(-1)。结论首次建立了复方黄芪乳膏HPLC指纹图谱结合含量测定的质量控制方法,为复方黄芪乳膏质量标准的建立和中药批次间的质量评价提供借鉴。
Objective To establish an HPLC fingerprints of Compound Huangqi cream and to simultaneously determine the content of 3 constituents.Methods The 75%methanol extract of Compound Huangqi cream was analyzed.Agilent 5 TC-C_(18)(4.6 mm×250 mm,5μm)chromatographic column was used.The mobile phase consisted of methanol-0.1%formic acid aqueous solution,flowing at 0.8 mL·min^(-1) with gradient elution.The detection wavelength was 250 nm.Results There were 18 common peaks in the fingerprints of 10 sample batches with the similarities over 0.930.According to the reference substance,geniponic acid,caffeic acid,hydroxysafflor yellow A,calycosin-7-glucoside,verbascoside and cinnamic acid were identified.The content of geniposide acid,calycosin-7-glucoside and cinnamic acid was at 1.59~1.84 mg·g^(-1),1.37~1.58 mg·g^(-1),and 0.791~0.943 mg·g^(-1),respectively.Conclusion The quality control method of Compound Huangqi cream with HPLC fingerprint is established and content determined for the first time,which can provide reference for the quality standard of Compound Huangqi cream and quality evaluation amongs Chinese medicine batches.
