基于肠道菌群探讨清肠栓治疗UC的作用机制 被引量:6

摘要 目的:观察清肠栓对两歧双歧杆菌、多形拟杆菌生长的影响,清肠栓干预后的粪菌对结肠炎模型大鼠Toll样受体4(TLR4)、核因子κB(NF-κB)表达的影响。方法:菌株复苏后,分别给予2.5%、5%、10%清肠栓组方煎剂、清肠栓组方煎剂原液和纯水培养,然后进行菌落计数。将24只雄性SD大鼠随机分为以下4组:正常组(N)、DSS造模组(M)、模型粪菌液灌肠+DSS造模组(M1)、清肠栓粪菌液灌肠+DSS造模组(M2)。研究采用5%DSS(w/v)造模7 d,造模同时给予N组和M组大鼠生理盐水灌肠,其余组大鼠给予相应粪菌液灌肠。计算动物疾病活动指数评分(disease active index DAI)和结肠组织学损伤(histological index,HI)评分,采用Western blot法测定结肠组织的TLR4/NF-κB表达。结果:与纯水培养组比较,清肠栓组方煎剂原液培养组的两歧双歧杆菌和多形拟杆菌的菌落数出现显著升高(P<0.01)。模型粪菌液灌肠+DSS造模组大鼠的HI评分较DSS组显著升高(P<0.05),清肠栓粪菌液灌肠+DSS造模组大鼠的HI评分较模型粪菌液灌肠+DSS造模组的显著下降(P<0.01)。清肠栓粪菌液灌肠+DSS造模组大鼠的TLR4/NF-κB表达较模型粪菌液灌肠+DSS造模组显著下降(P<0.05)。结论:清肠栓可促进两歧双歧杆菌和多形拟杆菌生长,调节UC菌群紊乱,抑制TLF4/NF-κB的表达,促进结肠黏膜的修复。 Objective:To observe the effect of Qingchang Supporitory on the growth of Bifidobacterium bifidum and Bacteroides thetaiotaomicron,and the effect of fecal bacteria on the expression of Toll-like receptor 4(TLR4)and nuclear factorκB(NF-κB)after the intervention of Qingchang Supporitory.Methods:Bifidobacterium bifidum and Bacteroides thetaiotaomicron were cultivated with water,2.5%,5.0%,10%and 100%Qingchang Supporitory,and the counts were compared.The model of ulcerative colitis(UC)was established by 5%Dextran Sulfate Sodium(DSS,w/v)for 7 days.24 SD rats were randomly divided into the normal group(N group),the DSS model group(M group),the model fecal enema plus DSS group(M1 group)and the Qingchang Supporitory fecal enema plus DSS group(M2 group).The scores of disease activity index(DAI)and histological index(HI)were calculated.The expression of TLR4/NF-κB in colon tissue was measured by Western blot method.Results:Bifidobacterium bifidum and Bacteroides thetaiotaomicron were significantly increased in the Qingchang Supporitory group compared to those in the pure water group(P<0.01).HI score was significantly increased in the M group compared to that in the M1 group(P<0.05);HI score was significantly decreased in M2 group compared to that in M1 group(P<0.01).The expression of TLR4/NF-κB was significantly reduced in M2 group compared to that in group M1(P<0.05).Conclusion:Qingchang Supporitory can promote the growth of Bifidobacterium bifidum and Bacteroides thetaiotaomicron,regulate flora disorder,suppress TLF4/NF-κB expression and improve the recovery of colonic muscosa in the treatment of UC.
