摘要
肽聚糖识别蛋白(peptidoglycan recognition protein, PGRP)是生物免疫系统中的一种模式识别受体,但在寄主昆虫响应寄生蜂寄生时发挥的免疫功能目前仍鲜有报道。本文旨在探究桑螟(Glyphodes pyloalis Walker)在响应混腔室茧蜂(Aulacocentrum confusum)寄生时GpPGRP发挥的免疫功能。本研究共鉴定得到3个短型GpPGRP编码基因,分别命名为GpPGRP-S1、GpPGRP-S2和GpPGRP-S3。GpPGRP-S1和GpPGRP-S2在雄性成虫阶段表达量最高,GpPGRP-S3在雌性成虫阶段表达量最高。与健康桑螟相比,3个GpPGRP的表达量在混腔室茧蜂寄生后的不同时间差异显著,且均在被寄生桑螟的血淋巴中上调表达。重组GpPGRP-S2蛋白显著提高健康桑螟血淋巴中酚氧化酶活性,表明桑螟GpPGRP可能通过增强血淋巴黑化反应响应混腔室茧蜂寄生。本研究为深入探究桑螟GpPGRP响应寄生蜂寄生时发挥的免疫功能提供了参考,为进一步揭示桑螟与寄生蜂之间的互作关系提供了依据。
Peptidoglycan recognition protein(PGRP) is a widely studied pattern recognition receptor in the immune system of organisms. However, the immune function of host insects in response to parasitic wasps is still rarely reported. The aim of this study was to investigate the immune function of GpPGRPs of Glyphodes pyloalis Walker in response to the parasitism by Aulacocentrum confusum. In this study, three short forms of GpPGRPs, namely GpPGRP-S1, GpPGRP-S2 and GpPGRP-S3, were identified. The expression level of GpPGRP-S1 and GpPGRP-S2 were the highest in male adult stage, and that of GpPGRP-S3 was the highest in female adult stage. Compared to healthy G. pyloalis, the expression levels of three GpPGRPs in parasitized G. pyloalis were significantly different at various time points after parasitization. Meanwhile, their expression levels were up-regulated in the hemolymph. Recombinant GpPGRP-S2 protein significantly enhanced the phenoloxidase activity in the hemolymph of the healthy G. pyloalis, suggesting that GpPGRP may respond to the parasitism of A. confusumby enhancing the melanization reaction in the hemolymph. This study provides a reference for further investigating the immune function of GpPGRPs in response to parasitic wasps and a basis for further study on the interaction between G. pyloalis and parasitic wasps.
