大鼠局灶性脑缺血再灌注血脑屏障通透性变化及对Occludin、ZO-1和PKCδ蛋白水平的影响 被引量:2

摘要 目的探索大鼠局灶性脑缺血再灌注血脑屏障通透性的变化及可能的相关机制。方法Wistar健康雄性大鼠60只,制备局灶性脑缺血模型,观察并比较脑缺血再灌注6 h、12 h、24 h、48 h的血脑屏障通透性及紧密连接蛋白(Occludin)、胞质紧密黏连蛋白1抗体(ZO-1)和蛋白激酶(PK)Cδ表达的差异。结果脑缺血再灌注后脑组织含水量明显高于空白对照组,且随时间而升高(P<0.05);脑缺血再灌注后脑组织伊文思蓝(EB)含量明显高于空白对照组,且随时间而升高(P<0.05);脑缺血再灌注后脑组织Occludin蛋白、ZO-1蛋白表达量明显低于空白对照组(P<0.05);脑缺血再灌注后脑组织PKCδ蛋白表达量明显高于空白对照组(P<0.05)。结论局灶性脑缺血再灌注后脑组织含水量和血脑屏障通透性增加,可能与Occludin蛋白低表达、ZO-1蛋白和PKCδ蛋白高表达有关。
