摘要
【目的】以红砂种子为试验材料,研究老化处理后红砂种子发芽指标和生理特性指标的变化规律,以期揭示红砂种子对高温高湿环境的响应机理.【方法】采用不同温度梯度(25(CK)、30、35、40、45、50℃)、不同处理时间(24、48、72 h)、相对湿度为95%的高湿环境对红砂(Reaumuria soongorica)种子进行人工老化处理.【结果】随着处理温度升高和时间延长,种子的发芽率呈先上升后下降的趋势.相对电导率呈增大趋势,显著高于对照19.2%~38.3%,保护酶活性呈先上升后下降的趋势,其中CAT与POD活性在处理温度30℃处理时间24 h时最高,并分别高出对照55.0%和94.1%,且与对照差异显著(P<0.05).而可溶性蛋白、可溶性糖、脯氨酸和MDA含量呈先升高后下降的趋势,其中,可溶性蛋白和可溶性糖分别较对照低17.6%~91.8%和87.0%~91.8%.【结论】老化处理使红砂种子细胞膜受损,造成渗透调节物质积累和保护酶系统受到破坏,老化时间24 h、温度30~40℃时红砂种子活力最佳.
【Objective】The study aimed to clarify the effect of aging on seed germination and physiological characteristics of Reaumuria soongorica,revealing the response mechanism of the seeds to the environment of high temperature and humidity.【Method】The artificial seed-aging treatment was carried out at the high relative humidity of 95%,with six temperature gradients(i.e.,25(CK),30,35,40,45 and 50℃)and three levels of treatment time(i.e.,24,48 and 72 h).【Result】With the increase of treatment temperature and time,the germination rate of the seeds increased at first and then decreased.An overall rising trend was observed for the relative conductivity of the seeds,which was increased by 19.2%~38.3%compared with the control.The activity of protective enzymes rose at first and then fell.Among them,both CAT and POD activities reached the highest point at 30℃for 24 hours,which increased by 55.0%and 94.1%,respectively,and significantly different with the control(P<0.05).The contents of soluble protein,soluble sugar,proline and MDA also increased at first and then declined,among which the contents of soluble protein and soluble sugar decreased by 17.6%~91.8%and 87.0%~91.8%,respectively,compared with the control.【Conclusion】Artificial aging could damage the cell membrane and protective enzyme system of R.soongorica seeds,resulting in the accumulation of osmotic regulation substances.The seed vigor was the highest when the aging was carried out at 30~40℃for 24 hours.
