摘要
目的探讨花椒毒酚对脊髓型颈椎病(CSM)大鼠的影响及其作用机制。方法将50只无特定病原级成年Sprague Dawley大鼠按随机数字表法分为假手术组(n=12)、模型组(n=13)、低剂量花椒毒酚组(n=12)和高剂量花椒毒酚组(n=13)。模型组、低剂量花椒毒酚组和高剂量花椒毒酚组大鼠于第4和第5颈椎椎间隙注入0.2 mL骨形态发生蛋白(BMP)混悬液制备CSM模型,假手术组大鼠注射0.2 mL生理盐水。造模后第3天,低剂量花椒毒酚组和高剂量花椒毒酚组大鼠分别给予腹腔注射5、10 mg·kg^(-1)花椒毒酚溶液,假手术组和模型组大鼠给予腹腔注射等量生理盐水,所有大鼠均隔日注射1次,连续注射7次。造模后第4周,采用苏木精-伊红染色法观察4组大鼠颈椎间盘滑膜组织病理学情况。采用改良Rivlin斜板实验观察4组大鼠造模前及造模后第4周的运动功能。采用酶联免疫吸附试验法检测4组大鼠血清及颈椎间盘关节液中白细胞介素(IL)-6、IL-1β、肿瘤坏死因子-α(TNF-α)水平。采用Western blot法检测4组大鼠颈椎间盘滑膜组织中NOD样受体蛋白3(NLRP3)、磷酸化核因子-κB p65(p-NF-κB p65)、核因子-κB(NF-κB)p65蛋白相对表达量,并计算p-NF-κB p65/NF-κB p65比值。结果造模后第4周,共3只大鼠死亡(假手术组1只、低剂量花椒毒酚组2只),其余大鼠的总体生长状态良好。模型组大鼠活动量略少,动作不如其他3组敏捷,呼吸幅度略显急促。假手术组大鼠的椎间盘滑膜组织结构完整,滑膜细胞排列整齐,未见显著炎症细胞浸润、充血及水肿;模型组大鼠可见滑膜组织结构显著增生、肿胀,表层细胞呈栅栏样排列,广泛存在炎症细胞浸润及脂肪组织填充;低剂量花椒毒酚组大鼠滑膜组织内炎症细胞浸润程度较模型组显著减轻,可见纤维化改变,间质内可见脂肪组织填充;高剂量花椒毒酚组未见明显炎症细胞浸润,增生及肿胀程度较低剂量花椒�
Objective To investigate the effect of xanthotoxin in the treatment of cervical spondylitis myelopathy(CSM)in rats and its mechanism.Methods Fifty adult Sprague Dawley rats with no specific pathogens were divided into sham operation group(n=12),model group(n=13),low-dose xanthoxylin group(n=12)and high-dose xanthoxylin group(n=13)according to the random number table method.The CSM models were prepared by injecting 0.2 mL of bone morphogenetic protein(BMP)suspension into the fourth and fifth cervical intervertebral spaces of rats in the model group,low-dose xanthoxylin group and high-dose xanthoxylin group.The rats in the sham operation group were injected with 0.2 mL normal saline.On the 3 rd day after modeling,rats in the low-dose xanthotoxin group and high-dose xanthotoxin group were given intraperitoneal injection of 5 mg·kg^(-1) and 10 mg·kg^(-1) xanthotoxin solution,respectively.The rats in the sham operation group and model group were injected intraperitoneally with the same amount of normal saline.All rats were injected once every other day for 7 consecutive injections.At four weeks after modeling,the histopathology of the synovial membrane of the cervical intervertebral disc of rats in the four groups was observed by hematoxylin-eosin staining.The motor function of rats in the four groups was observed by the modified Rivlin oblique plate experiment before and four weeks after modeling.The levels of interleukin(IL)-6,IL-1β,and tumor necrosis factor-α(TNF-α)in serum and cervical intervertebral disc synovial fluid of rats in the four groups were detected by the enzyme-linked immunosorbent assay method.The relative expression levels of NOD-like receptor protein 3(NLRP3),phosphorylated nuclear factor-κB p65(p-NF-κB p65)and nuclear factor-κB(NF-κB)p65 in the synovial tissues of cervical intervertebral discs of rats in the four groups were detected by Western blot,and the ratio of p-NF-κB p65/NF-κB p65 was calculated.Results At four weeks after modeling,three rats died in total(one rat in the sham ope
