甜橙UDP-糖基转移酶基因的筛选及其功能分析 被引量:1

Screening and functional analysis of UDP-glycosyltransferase genes in sweet orange(Citrus sinensis)

摘要 UDP-糖基转移酶在植物中通常以基因家族的形式存在,在进行植物糖基化研究时,常常难以克隆到所需的目的基因。本研究对甜橙子房、花后20 d及40 d幼果进行转录组测序,筛选出了36个高表达Cs UGT基因。36个CsUGT基因的编码蛋白在motif组成上高度相似,其中,36个CsUGT的C端普遍含有motif 1和motif 2,24个Cs UGT的N端含有motif 3。它们的氨基酸数介于345~780 aa之间,分子量介于38.52~87.55kDa之间,等电点介于4.76~7.93之间。亚细胞定位预测显示,16个CsUGT定位于细胞质,4个定位于叶绿体,1个定位于胞外基质,9个定位于细胞质膜,2个可能定位于细胞质或叶绿体,4个可能定位在细胞质膜或细胞质。进一步的进化聚类分析发现,有6个CsUGT基因的编码蛋白可能参与了激素的糖基化修饰,20个可能参与了类黄酮的糖基化修饰。 Plant UGTs are commonly composed of a large gene family. Thus, it is difficult to identify key genes from a huge number of family numbers. In this study, transcriptomes were performed on ovary, fruits at 20 and 40 days after blooming to screen UGT genes with higher expression level. A total of 36 Cs UGT genes with higher expression were screened. The encoded proteins of 36 Cs UGT genes were highly similar in motif composition. Among them, the C-terminal of 36 CsUGTs generally contained motif 1 and motif 2, and the N-terminal of 24 CsUGTs contained motif 3. Their amino acid numbers were between 345–780 aa, their molecular weights were between 38.52–87.55 kDa, and their isoelectric points were between 4.76–7.93. The prediction of subcellular location showed that 16 CsUGTs were located in the cytoplasm, 4 were located in the chloroplast, 1 was located in the extracellular matrix, 9 were located in the plasma membrane, 2 were located in the cytoplasm or chloroplast, and 4 were located in the plasma membrane or cytoplasm. Among them, 6 UGTs might be involved in the glycosylation of plant hormones, while 20 might function in the flavonoid glycosylation.
