摘要
葡萄(Vitis vinifera)咖啡酸-O-甲基转移酶(COMT)基因存在三个可变剪切本,本文主要研究了VvCOMT(XP019071814.1)的表达特性及其在褪黑素和阿魏酸合成上的功能。结果表明,VvCOMT与番茄和拟南芥COMT同源性在80%以上。VvCOMT在果肉和种子表达量较高,果实成熟过程中,VvCOMT在果皮中表达量逐渐下降,在种子中呈上升趋势。在果实成熟过程中,VvCOMT表达水平与褪黑素含量极显著正相关。在拟南芥叶片和葡萄愈伤组织中过量表达VvCOMT,均能促进褪黑素和阿魏酸合成,并且以促进褪黑素合成为主。同时发现,转基因材料中阿魏酸增幅与褪黑素增幅相反。进一步外源褪黑素处理表明,褪黑素含量升高能抑制阿魏酸合成。
There exist three alternative splicing transcripts of caffeic acid-O-methyltransferase(COMT) in grape(Vitis vinifera), and the expression characteristics and functions of VvCOMT(XP019071814.1) in synthesizing melatonin and ferulic acid were investigated in this study. The results showed that VvCOMT has more than 80% sequence similarity with its counterparts from tomato and Arabidopsis. The expression of Vv COMT was higher in pulp and seeds, which gradually decreased in peel while showed the increasing trend in seeds during ripening. Additionally, the expression level of Vv COMT was very significantly positively correlated to melatonin content during ripening. Overexpression of VvCOMT in Arabidopsis leaves and grape calluses increased the content of ferulic acid and particularly melatonin. Additionally, the increment of ferulic acid showed the contrary changes to that of melatonin. Further treatment of exogenous melatonin demonstrated that the increase in melatonin content inhibited the biosynthesis of ferulic acid.
