摘要
目的基于干细胞因子(stem cell factor,SCF)/c-kit受体复合物对胃窦Cajal间质细胞(interstitial cells of Cajal,ICC)数量的调控作用,研究中焦点穴对功能性消化不良(functional dyspepsia,FD)大鼠胃动力调节的机制。方法36只大鼠随机分为空白组、模型组、西药组、点穴组、阻断点穴组、模型阻断组,每组6只。除空白组外,其余各组采用轻度夹尾刺激+不规则饮食法持续造模21天。阻断点穴组、模型阻断组大鼠于造模第12天起,予以Kit单克隆抗体(ACK2)腹腔注射(0.2 mg/kg),隔日1次,持续26天。造模成功后点穴组与阻断点穴组拨揉阑门、建里穴,每日1次,连续14天;药物组以10 mL/kg莫沙必利悬浊液灌胃,每日1次,连续14天。干预结束后,检测各组大鼠胃内残留率;免疫组化检测胃窦ANO1积分光密度值(IOD值)、阳性面积比;Western Blot法检测胃窦SCF、c-kit、磷酯酰肌醇-3-激酶(phosphatidylinositol 3-kinases,PI3K)蛋白表达。结果模型组大鼠胃内残留率高于空白组(P<0.05);点穴组、西药组胃内残留率低于模型组(P<0.05);点穴组胃内残留率与西药组比较,差异无统计学意义(P>0.05);阻断点穴组、模型阻断组胃内残留率高于点穴组(P<0.05);阻断点穴组胃内残留率略低于模型阻断组,差异无统计学意义(P>0.05)。模型组胃窦SCF、c-kit、PI3K表达与IOD值、阳性面积比低于空白组(P<0.05);点穴组、西药组胃窦SCF、c-kit、PI3K表达与IOD值、阳性面积比高于模型组(P<0.05);阻断点穴组胃窦c-kit、PI3K表达与IOD值、阳性面积比低于点穴组(P<0.05),SCF表达低于点穴组,差异无统计学意义(P>0.05);阻断点穴组胃窦c-kit、PI3K表达与IOD值、阳性面积比略高于模型阻断组,差异无统计学意义(P>0.05),SCF表达高于模型阻断组(P<0.05)。结论(1)中焦点穴可促进FD大鼠胃动力恢复;(2)SCF/c-kit通路是中焦点穴促胃动力恢复的重要机制之一;(3)中焦点穴或可增加ICC数量,并可能通�
Objective To study the regulation mechanism of middle energizer point on gastric motility in rats based on the regulation effect of SCF(Stem cell factor)/c-kit on the quantity of ICC(Interstitial cells of Cajal).Methods 36 rats were randomly divided into normal group,model group,medicine group,point pressing group,blocking point pressing group and blocking point pressing group,6 rats in each group.In addition to the normal group,the other groups were treated with mild tail-clamping stimulation plus irregular diet for 21 days.After the model established successfully,the rats in point pressing group and blocking point pressing group were kneading-puling the points of Lanmen and Jianli once a day for 14 days;the rats in the medicine group were treated with 10 mL/kg mosapride suspension by gavage once a day for 14 days.The rats in the blocking point group and the blocking model group were given ACK2 intraperitoneal injection(0.2 mg/kg)from the 12th day of modeling,once every other day,lasting for 26 days.After the intervention,the intragastric residual rate of rats in each group were detected;integrated optical density(IOD)of ANO1 and positive area percentage in antrum was detected by immunohistochemistry;the expression of SCF,c-kit and PI3K in antrum were detected by Western blot.Results The gastric residual rate in the model group was higher than that in the normal group(P<0.05);the gastric residual rate in the point pressing group and the medicine group were lower than that in the model group(P<0.05);the intragastric residual rate in the point pressing group was not significantly different from that in the medicine group(P>0.05);the gastric residual rate in the blocking point pressing group and the blocking model group was higher than that in the point pressing group(P<0.05);the gastric residual rate in the blocking point pressing group was slightly lower than that in the blocking model group,the difference was not statistically significant(P>0.05).The IOD of ANO1,positive area percentage and expression of SCF,c-k
