摘要
以两种益生元低聚果糖(FOS)、低聚异麦芽糖(IMO)作为培养基碳源,以无糖培养基和普通MRS培养基为阴性和阳性对照,考察低聚糖益生元对干酪乳杆菌(L.casei)增殖作用的影响。结果表明,FOS和IMO均能促进L.casei的增殖,且FOS的益生效果优于IMO。通过在无糖培养基中添加不同浓度的FOS作为碳源,进一步探究其对益生菌增殖的影响规律,结果表明,L.casei的增殖随FOS浓度的增大而逐渐提高,当FOS浓度>2%时,继续增加浓度对菌的增殖无明显促进作用。该实验内容具有开放性和探索性,把传统的“以课程为中心”的实验教学切换为“以问题为中心”的科研性教学。该文从教学案例选择、实验项目实施过程及实验结果讨论等方面总结了实验项目在本科教学中的应用,为同类课程教学改革提供参考。
Taking two prebiotics fructooligosaccharides(FOS)and isomaltooligosaccharides(IMO)as carbon source of culture medium,sugar free medium and ordinary MRS medium as negative and positive controls,effect of oligosaccharide prebiotics on proliferation of L.casei was investigate.Results showed that both FOS and IMO could promote proliferation of L.casei,and prebiotic effect of FOS was better than IMO.Effect of FOS on proliferation of probiotics was further explored by adding different concentrations of FOS as carbon source in sugar free medium.Results showed that proliferation of L.casei increased gradually with increase of FOS concentration.When FOS concentration was higher than 2%,continuous increase of concentration did not significantly promote proliferation of L.casei.This experimental content was open and exploratory.And it switched traditional“curriculum centered”experimental teaching to“problem centered”scientific research teaching.Application of experimental items in undergraduate teaching was summarized from selection of teaching cases,implementation process of experimental projects and discussion of experimental results.It provided reference for teaching reform of similar courses.
