摘要
目的:探讨延龄草总皂苷对肺癌A549细胞增殖、迁移和侵袭的影响以及其对miR-1470的调控作用。方法:体外人肺癌细胞A549、延龄草总皂苷处理细胞、anti-miR-NC、anti-miR-1470分别转染入A549细胞,miR-NC、miR-1470 mimics分别转染入A549细胞后加入延龄草总皂苷处理细胞;采用MTT实验检测细胞增殖抑制率;采用Transwell实验检测细胞迁移及侵袭;采用qRT-PCR法检测miR-1470的表达量。结果:延龄草总皂苷处理后细胞增殖抑制率升高(P<0.05),迁移及侵袭细胞数均明显减少(P<0.05),miR-1470的表达水平降低(P<0.05);转染anti-miR-1470后细胞增殖抑制率升高(P<0.05),迁移及侵袭细胞数均明显减少(P<0.05)。结论:延龄草总皂苷可通过下调miR-1470的表达减弱肺癌细胞增殖、迁移及侵袭能力。
Objective:To explore the effect of trillium saponins on the proliferation,migration and invasion of lung cancer A549 cells and its regulation of miR-1470.Methods:In vitro human lung cancer cells A549,total saponins of trillium treated cells,anti-miR-NC and anti-miR-1470 were transfected into A549 cells respectively,and miR-NC and miR-1470 mimics were transfected into A549 cells respectively and then the total saponins of trillium treated cells were added.MTT assay was used to detect the inhibition rate of cell proliferation.Transwell test was used to detect cell migration and invasion.The expression of miR-1470 was detected by qRT-PCR.Results:After trillium saponins treatment,the inhibition rate of cell proliferation was signifi cantly increased(P<0.05).The migration and the number of invasive cells were signifi cantly reduced(P<0.05),and the expression of miR-1470 was signifi cantly reduced(P<0.05).After transfection with anti-miR-1470,the inhibition rate of cell proliferation was signifi cantly increased(P<0.05)and the number of migration and invasive cells were signifi cantly reduced(P<0.05).Conclusion:The total saponins of trillium could reduce the proliferation,migration and invasion of lung cancer cells by down-regulating the expression of miR-1470.
