摘要
以甘草为原料,采用超声辅助乙醇提取法提取其多酚类物质,通过单因素试验和响应面法优化提取工艺,利用Fenton反应法、邻苯三酚自氧化法以及1,1-二苯基-2-三硝基苯肼(DPPH)比色法分析甘草多酚对羟基自由基(·OH)、超氧阴离子(O_(2)^(–)·)和DPPH自由基的清除能力。结果表明,甘草多酚的最佳提取工艺参数为:乙醇浓度70%、料液比1:42 g/mL、超声时间66 min、超声功率250 W,该条件下多酚提取率为4.97%。抗氧化试验表明,0.5 mg/mL的甘草多酚对·OH、O_(2)^(–)·和DPPH·的清除率分别为78.83%、72.15%、90.33%。试验结果为甘草多酚的有效提取及甘草的深度开发利用提供理论依据。
The polyphenols were extracted from the Glycyrrhiza by the ultrasound-assisted ethanol extraction method.The extraction process was optimized by single factor test and response surface method.Fenton reaction method,pyrogallol autooxidation method,and 1,1-diphenyl-2-trinitrophenylhydrazine(DPPH)colorimetric method were used to determine the scavenging ability of Glycyrrhiza polyphenols to hydroxyl radical(·OH),superoxide anion(O_(2)^(–)·),and DPPH·,respectively.The results showed that the optimum extraction parameters of Glycyrrhiza polyphenols were ethanol concentration 70%,material-liquid ratio 1:42 g/mL,ultrasound time 66 min,and ultrasonic power 250 W.The extraction rate of polyphenols was 4.97%under the optimal condition.The antioxidant test showed that the scavenging rates of 0.5 mg/mL Glycyrrhiza polyphenols to·OH,O_(2)^(–)·and DPPH·were 78.83%,72.15%,and 90.33%,respectively.These results provided a theoretical basis for the effective extraction of polyphenols from Glycyrrhiza and the further development and utilization of Glycyrrhiza.
