摘要
目的研究固本化瘀法对2型糖尿病大鼠足细胞损伤及磷脂酰肌醇3-激酶/蛋白激酶B(PI3K/Akt)信号通路的影响。方法选择成年雄性SD大鼠并随机分为对照组,模型组,固本化瘀方低、中和高剂量组,采用高脂高糖饲料喂养联合链脲佐菌素腹腔注射的方式建立2型糖尿病模型,固本化瘀方低、中、高剂量组分别给予22.1、44.2、88.4 g·kg^(-1)固本化瘀方灌胃,连续4周,检测24 h尿微量白蛋白(mAlb)/尿肌酐(Ucr)、空腹血糖(FBG)、血肌酐(Scr)和血尿素氮(BUN)含量及肾脏病理改变、肾病蛋白(nephrin)、足细胞蛋白(podocin)、CD2相关蛋白(CD2AP)、磷酸化磷脂酰肌醇3-激酶(p-PI3K)和磷酸化蛋白激酶B(p-AKT)的表达水平。结果模型组大鼠的FBG、Scr、BUN、尿mAlb/Ucr均高于对照组,肾脏组织中nephrin、podocin、CD2AP、p-PI3K、p-Akt的表达水平均低于对照组;固本化瘀方低、中、高剂量组的FBG、Scr、BUN、尿mAlb/Ucr均低于模型组,肾脏组织中nephrin、podocin、CD2AP、p-PI3K和p-Akt的表达水平均高于模型组。结论固本化瘀法可减轻2型糖尿病大鼠足细胞损伤,该作用与激活PI3K/Akt通路有关。
Objective To study the effect of consolidating the root and resolving blood stasis method on podocyte injury and phosphatidylinositol 3-kinase and protein kinase B(PI3K/Akt)signaling pathway in type 2 diabetic rats.Methods Adult male SD rats were randomly divided into control group,model group,consolidating the root and resolving blood stasis prescription low-dose group,medium dose group and high-dose group.The model of type 2 diabetic was established by feeding high-fat and high sugar diet combined with intraperitoneal injection of streptozotocin.Consolidating the root and resolving blood stasis prescription was given by gavage at 22.1,44.2 and 88.4 g·kg^(-1) for 4 weeks.The fasting blood glucose(FBG),serum creatinine(Scr),blood urea nitrogen(BUN)and renal pathological changes,nephrin,podocin,CD2AP,p-PI3K,p-Akt expression levels were detected.Results FBG,Scr,BUN and urine mAlb/UCR of model group were higher than those of control group,and the expression levels of nephrin,podocin,CD2AP,p-PI3K and p-Akt in kidney tissue were lower than those of control group;FBG,Scr,BUN and urine mAlb/Ucr of low-dose group,middle dose group and high-dose group were lower than those of model group,and the expression levels of nephrin,podocin,CD2AP,p-PI3K and p-Akt in kidney tissue were higher than those of model group.Conclusion Consolidating the root and resolving blood stasis method can alleviate podocyte injury in type 2 diabetic rats,which relates to the activation of PI3K/Akt pathway.
