摘要
为了明确施用宁南霉素和生防菌对烟草内生菌群落的影响,采用宁南霉素和烟草内生解淀粉芽孢杆菌YN201732(Y32)不同先后喷施组合,提取处理后不同时间段烟草叶片,运用高通量测序技术测定内生细菌16S rDNA序列,分析菌群多样性。结果表明,仅喷施Y32发酵液(10^(7) cfu/mL)的叶片内细菌物种多样性高于其他处理。先喷施Y32发酵液,24 h后再喷施宁南霉素的叶片内细菌物种丰富度高于先喷施宁南霉素,24 h后再喷施Y32发酵液的处理。所有样品中优势细菌属均为泛菌属、沙雷氏菌属、芽孢杆菌属和假单孢菌属,它们的相对丰度之和大于80%。Y32发酵液能有效提高烟草叶片新陈代谢等生命活动,加快生命进程。宁南霉素能降低烟草内生细菌物种多样性,而烟草内生细菌Y32能够提高烟草叶片内生细菌物种多样性,这一结果对利用内生菌开展生物防治具有重要指导意义。
In order to clarify the effects of ningnanmycin and biocontrol bacteria on endophytic bacterial community of tobacco plants,the leaves of tobacco were extracted by spraying ningnanmycin and Bacillus amyloliquefaciens YN201732(Y32)at different times.The 16S rDNA sequence of endophytic bacteria was determined by high-throughput sequencing technology,and the diversity of bacterial community was analyzed.The results indicated that the bacterial species diversity in leaves sprayed with Y32 fermentation broth(10^(7) cfu/mL)was higher than that in other treatments.Higher species richness were observed in the tobacco leaves when fermented suspension of strain Y32 was sprayed first,followed by ningnanmycin after 24 hours.In addition,ningnanmycin applied to tobacco leaves initially,followed by Y32 after 24 hours.The dominant bacterial genera in all samples were Pantoea,Serratia,Bacillus and Pseudomonas,and the sum of their relative abundance was more than 80%.Y32 fermented suspension could effectively improve metabolism and speed up the life process of tobacco leaves.The application of ningnanmycin resulted in the number reduction of endophytic bacterial communities,while strain Y32 could increase the endophytic bacteria in tobacco leaves.These results have important guiding significance for biocontrol by endophytic bacteria.
