摘要
目的探寻meso取代的氟硼二吡咯作为光敏剂的潜能,考察其光物理性质和在体内外的光动力抗肿瘤性能。方法经缩合反应制备氟硼二吡咯类光敏剂(BOD-OH),并采用核磁共振氢谱、碳谱和红外吸收光谱对其进行结构表征。通过吸收光谱法和荧光光谱法考察其光物理性质;运用DPBF法考察其体外光辐射产生单线态氧的性能;使用荧光成像法和MTT法考察4T1(小鼠乳腺癌细胞)对BOD-OH的细胞摄取性能及其对4T1细胞的光动力学性能;通过瘤内注射考察BOD-OH在4T1肿瘤模型的BALB/c小鼠体内光动力抗肿瘤性能。结果核磁共振氢谱、碳谱和傅里叶红外光谱证实合成的BOD-OH为预期目标分子;体外实验结果表明,BOD-OH具有良好的光物理性能、较高的单线态氧产生能力、良好的4T1细胞摄取性能及优异的光动力学性能;体内实验结果表明,BOD-OH具有良好的4T1肿瘤模型光动力治疗性能。结论获得了一个具有较好应用前景的氟硼二吡咯类光敏剂,为氟硼二吡咯类作为光敏剂研究奠定了实验基础。
Objective To develop the potential of meso-substituted boron dipyrromethene as photosensitizers,and evaluate their photophysical properties and photodynamic effects in vivo and invitro.Methods The boron dipyrromethene photosensitizer BOD-OH was prepared by condensation reaction,and its structure was characterized by hydrogen nuclear magnetic resonance spectrum(^(1)H-NMR),carbon nuclear magnetic resonance spectrum(13 C-NMR)and fourier transform infrared spectroscopy(FT-IR).The photophysical properties of BOD-OH were tested by absorption spectroscopy and fluorescence spectroscopy.DPBF method was used to investigate the ^(1)O_(2) performance of BOD-OH in vitro under light radiation.MTT method was used to investigate the cellular uptake of BOD-OH by 4T1(mouse breast cancer cells)and its photodynamic therapy effects on 4T1 cells.With the commercially available photosensitizer Ce6 and chlorambucil as the positive control and negative control,the photodynamic anti-tumor effects of BOD-OH on the 4T1 tumor model of BALB/c mice were studies.Results Proton nuclear magnetic resonance spectroscopy(^(1)H-NMR),carbon nuclear magnetic resonance spectroscopy(13 C-NMR)and Fourier transform infrared spectroscopy(FT-IR)confirmed that the synthesized BOD-OH was the expected structure.In vitro experimental results showed that BOD-OH had good photophysical properties,high singlet oxygen generation capacity,superior fluorescence performance of 4T1 cells and excellent photodynamic effect.In vivo experimental results showed that BOD-OH had good photodynamic therapy effect than Ce6 and chlorambucil.Conclusion A boron dipyrromethene photosensitizer with good application prospects has been obtained,which would be a reliable foundation for the study of the boron dipyrromethene photosensitizeras photosensitizers.
