摘要
目的:采用网络药理学的方法分析芍药甘草附子汤医治类风湿性关节炎可能的作用机制。方法:利用中药系统药理学分析平台,以口服生物利用度(OB)≥30%、类药性(DL)≥0.18作为指标,挑选芍药甘草附子汤的药效成分及作用靶点,构建化合物-靶点网络。利用DisGeNET和TTD数据库挑选类风湿关节炎有关的作用靶点,利用String数据库建立蛋白质相互作用网络,利用软件RStudio中ClusterProfiler包进行基因本体(GO)和基因功能(KEGG)富集分析。结果:芍药甘草附子汤中筛选出94个活性成分,涉及治疗类风湿性关节炎的113个靶点,该方可能通过影响IL-17信号通路、TNF信号通路、PI3K-Akt信号通路、HIF-1信号通路、Toll样受体信号通路、JAK-STAT信号通路、T细胞受体信号通路等发挥治疗风湿性关节炎的作用。结论:芍药甘草附子汤治疗类风湿性关节炎具有多成分、多靶点、多途径的特点。
Objective The possible mechanism of Shaoyao Gancao Fuzi Decoction in the treatment of rheumatoid arthritis was analyzed by network pharmacology.Methods Using the pharmacological analysis platform of traditional Chinese medi⁃cine system,taking oral bioavailability(OB)≥30%and drug class(DL)≥0.18 as indicators,the pharmacodynamic com⁃ponents and action targets of Shaoyao Gancao Fuzi Decoction were selected to construct the compound target network.The targets related to rheumatoid arthritis were selected by using disgenet and TTD database,the protein interaction network was established by using string database,and gene ontology(GO)and gene function(KEGG)enrichment analysis were per⁃formed using ClusterProfiler package in software RStudio.Results 94 active ingredients were screened from Shaoyao Gancao Fuzi decoction,involving 113 targets for the treatment of rheumatoid arthritis.This prescription may play a role in the treat⁃ment of rheumatoid arthritis by affecting IL-17 signal pathway,TNF signal pathway,PI3k-Akt signal pathway,HIF-1 sig⁃nal pathway,Toll like receptor signal pathway,JAK-STAT signal pathway and T cell receptor signal pathway.Conclusion Shaoyao Gancao Fuzi Decoction has the characteristics of multiple components,multiple targets and multiple pathways in the treatment of rheumatoid arthritis.
