摘要
Keap1-NRF2系统是细胞针对活性氧(ROS)和亲电基团所引起的内外源性应激反应的重要调节机制。虽然转录因子NRF2的短暂激活阻止了正常组织和癌前组织中癌症的发生发展,但在恶性肿瘤细胞中NRF2的持续激活通过增加肿瘤治疗耐药性和促进肿瘤细胞生长从而对宿主产生有害影响。本综述对Keap1-NRF2在癌症中的激活与失调机制进行总结并探索其在癌症基因治疗中的应用。
The Keap1-NRF2 system is the master orchestrator of the cellular responses to endogenous and exogenous stress induced by reactive oxygen species(ROS)and electrophiles.While transient activation of transcription factor NRF2 prevents cancer development in normal and precancerous tissues,persistent activation of NRF2 in malignant tumor cells has deleterious effects on the host by increasing resistance and promoting tumor cell growth.This review sums up the mechanisms of Keap1-NRF2 activation and dysregulation in cancer and explores its application in cancer gene therapy by researching the consequences of them on cancer cells.
