摘要
为加强岷县黑裘皮羊及贵德黑裘皮羊种质资源的合理利用及保护,本研究对6个岷县黑裘皮羊样本以及5个贵德黑裘皮羊样本线粒体DNA的Cytb、COI、ND2、ND4四对线粒体基因序列进行PCR扩增及序列分析。结果表明:岷县黑裘皮羊与贵德黑裘皮羊之间、岷县黑裘皮羊与国内外绵羊品种之间以及贵德黑裘皮羊与国内外绵羊品种之间均存在一定的基因流。其中,岷县黑裘皮羊的样本M1与样本M4受到外来杂交品种基因的冲击较大。基于上述结果,本研究对于岷县及贵德黑裘皮羊提出了保种建议:一是避免岷县及贵德黑裘皮羊与其他绵羊品种产生基因流;二是加强岷县及贵德黑裘皮羊的品种提纯工作。
In order to strengthen the rational utilization and protection of germplasm resources of Minxian black fur sheep and Guide black fur sheep,PCR amplification and sequence analysis of Cytb,COI,ND2 and ND4 mitochondrial gene sequences of mitochondrial DNA of 6 Minxian black fur sheep samples and 5 Guide black fur sheep samples were carried out.The results showed that there were certain gene flows between Minxian black fur sheep and Guide black fur sheep,between Minxian black fur sheep and domestic and foreign sheep breeds,and between Guide black fur sheep and domestic and foreign sheep breeds.Among them,samples M1 and M4 of Minxian black fur sheep were greatly impacted by the genes of foreign hybrids.Based on the above results,the suggestions for the conservation of Minxian and Guide black fur sheep are as follows:first,avoid gene flow between Minxian and Guide black fur sheep and other sheep breeds;Second,strengthen the variety purification of Minxian and Guide black fur sheep.
