摘要
目的探讨氯膦酸二钠脂质体(CL)治疗膝关节骨性关节炎(KOA)的效果。方法30只小鼠均进行KOA造模,造模成功后随机分为3组,每组各10只,分别行膝关节腔注射CL、帕瑞西布钠和生理盐水,4周及8周后获取膝关节标本,观察膝关节病理变化。体外培养巨噬细胞系,分别加入CL、空白脂质体,观察其对巨噬细胞的影响。结果CL组小鼠膝关节病理切片可见软骨退变较少,关节表面破坏较轻。不同浓度CL培养下的巨噬细胞存在不同程度的凋亡,高浓度下巨噬细胞凋亡明显,低浓度下反而对巨噬细胞增殖有促进作用。结论CL可以诱导巨噬细胞凋亡,在一定程度上减少软骨细胞的破坏,保护软骨细胞,以减缓关节的退变。
Objective To investigate the effect of clodronate liposomes(CL)to treat knee osteoarthritis(KOA).Methods 30 mice were randomly divided into 3 groups and all performed knee osteoarthritis modeling.After the modeling was successful,the knee joint cavity was injected with clodronate liposome,Parecoxib sodium and saline.Knee joint specimens were taken 4 week and 8 weeks to observe the pathological changes of the knee joint.The macrophage cell was cultured in vitro,and clodronate liposomes and control liposomes were added to observe their effects on macrophages.Results The pathological section of the knee joint of mice in the clodronate liposome group showed less cartilage degeneration and less damage to the joint surface,macrophages cultured in different concentrations of clodronate liposomes had different degrees of apoptosis,high apoptosis of macrophages was obvious at high concentration,but at low concentration,it promoted the proliferation of macrophages.Conclusion Clodronate liposomes can induce apoptosis of macrophages,reduce the destruction of chondrocytes to a certain extent,and protect cartilage cells to slow down the degeneration of joints.
