摘要
用分光光度法研究了37℃、pH=5.5、0.1M NaClO4介质中cis[Pt(NH_(3))_(2)Cl_(2)]和DNA组成物——鸟嘌呤核苷、腺嘌呤核苷、胞嘧啶核苷及胸腺嘧啶核苷的作用。发现顺-[PtⅡ(NH_(3))_(2)]与前三种核苷能生成组成为1:1、1:2二种络合物,与胸腺嘧啶核苷不作用。所测得一级和二级表观生成常数,以及作用初速分别有如下大小次序:Guo>Ado>Cyt>>Thy;Guo>Ado>Cyt>>Thy.在所得结果基础上讨论了顺-[Pt(NH_(3))_(2)Cl_(2)]和癌细胞中DNA作用的可能方式。
The interaction of cis-[Pt(NH_(3))_(2)Cl_(2)]with guanosine,adenosine,cyti-dine and thymidine,as constituents of DNA,was studied spectrophotome-trically at pH5.5,37°C in the presence of 0.1M NaClO_(4).It was found that the first three nucleosides formed 1:1 and 1:2 complexes with cis-CptⅡ(NH_(3))_(2)],whose apparent stability constants and initial rate of formation are given as follows:■Thymidine showed no interaction with cis-[PtⅡ(NH_(3))_(2)].On the basis of the above findings,a possible mode of reaction of cis-[Pt(NH_(3))_(2)Cl_(2)]with DNA in the cancer cells is discussed.Thermodynami-cally and kinetically,it is considered most probable that cross links were formed between cis-[PtⅡ(NH_(3))_(2)]and nucleosides in DNA,e.g.,guanosine,and it seems unlikely that a chelate was formed withany one nucleoside,e.g.,■.
