摘要
目的:观察穿龙薯蓣皂苷对再生障碍性贫血(AA)小鼠血管内皮生长因子(VEGF)和表皮生长因子受体1(EGFR1)表达的影响,探究其在骨髓造血微环境中发挥作用的可能机制。方法:40只BALB/c小鼠随机分为正常组和AA模型组,AA模型组经137Csγ射线全身照射后,于第4、5、6天腹腔注射环磷酰胺(25mg·kg^(-1)·d^(-1))和氯霉素(62.5mg·kg^(-1)·d^(-1)),建立AA小鼠模型。第7天模型评估后,将成模小鼠随机分为模型组、穿龙薯蓣皂苷组(DNS组)和环孢素A组(CsA组),进行14d灌胃干预。第22天,检测各组白细胞(WBC)、血红蛋白(HGB)、血小板(PLT)及网织红细胞(RET),采用瑞氏-姬姆萨染色法观察小鼠骨髓增生情况,采用Western blot法检测小鼠骨髓细胞VEGF、EGFR1蛋白的表达。结果:与正常组比较,模型组WBC、HGB、PLT、RET均显著减低(P<0.01),VEGF、EGFR1蛋白表达均减少(P<0.01);与模型组比较,DNS组和CsA组WBC、HGB、PLT、RET显著升高(P<0.01),VEGF、EGFR1蛋白表达显著增加(P<0.01),且DNS组VEGF、EGFR1蛋白高于CsA组(P<0.01)。结论:穿龙薯蓣皂苷通过促进VEGF、EGFR1的表达,增加血管新生,改善造血微环境,恢复AA小鼠骨髓造血。
Objective: To observe the effects of Dioscorea nipponica saponins(DNS) on the expressions of VEGF and EGFR1 in AA mice, and explore the possible mechanism of its role in the bone marrow hematopoietic microenvironment. Methods: Forty BALB/c mice were randomly divided into normal group and AA model group. The mice in AA model group were irradiated with 137 Cs γ-rays and intraperitoneally injected with cyclophosphamide(25 mg·kg^(-1)·d^(-1)) and chloramphenicol(62.5 mg·kg^(-1)·d^(-1)) on the 4 th, 5 th and 6 th day. The model was evaluated on day 7. After model evaluation, the model mice were randomly divided into model group, DNS group and cyclosporine A(CsA) group, and they were given intragastric intervention for 14 days. On the 22 nd day, the counts of WBC, HGB,PLT and RET in each group were detected, the bone marrow hyperplasia was observed by Wright-Giemsa staining and the expressions of VEGF and EGFR1 proteins in bone marrow cells were detected by Western blot. Results: Compared with the normal group, the counts of WBC, HGB, PLT and RET in model group significantly decreased(P<0.01), and the expressions of VEGF and EGFR1 in model group obviously reduced(P<0.01). Compared with the AA model group, the counts of WBC, HGB, PLT and RET significantly increased(P<0.01) and the expressions of VEGF and EGFR1 increased(P<0.01) in DNS group and CsA group, the expressions of VEGF and EGFR1 in DSN group were higher than those in CsA group(P<0.01). Conclusion: By promoting the expressions of VEGF and EGFR1, DNS can increase angiogenesis, improve the hematopoietic microenvironment and restore bone marrow hematopoiesis in AA mice.
