摘要
目的:利用稳定同位素比率及微量元素含量差异,结合化学计量学模型,探讨当归产地溯源应用的可行性,以期为当归产地识别提供理论支撑。方法:采用稳定同位素质谱仪(EA⁃IRMS)测定来自甘肃(岷归)、云南(云归)、四川(川归)3个主产区,共计27批次的当归样品中δ^(13)C、δ^(15)N稳定同位素比例以及碳、氮元素百分含量,通过电感耦合等离子体质谱仪(ICP⁃MS)测定样品中7种微量元素(锰、锌、铬、镁、铁、镍、钼)的含量。采用逐步线性判别分析法(FLDA)进行多变量判别分析。结果:不同产区当归中的δ^(13)C、δ^(15)N稳定同位素比率表现出产地差异;当归中镁、铬、铁元素含量较丰富,四川当归中钼元素含量较高,云南产地当归中锰、镁、铁、镍、锌元素含量丰富。稳定同位素及C%、N%作为判别变量的FLDA经回代检验和交叉验证,产地判别准确率达90%和83%,对外部样品预测准确度为63%,对大产地(省级行政区域)区分度较好,对小产地(县市级行政区域)的区分度不高;增加微量元素作为判别变量后,FLDA回代正确率达100%,交叉验证正确率达91%,对外部样品预测准确率100%。结论:当归中δ^(13)C、δ^(15)N稳定同位素、C%、N%及微量元素带有明显的地域特征,利用矿物元素和稳定同位素相结合的技术可以对当归进行有效的产地判别,判别正确率较高,是产地溯源的有效方法。
Objective:To explore the feasibility of tracing the origin of Angelicae Sinensis Radix by using stable isotope ratio and trace element content in combination with chemometric models,so as to provide theoretical support for the production area recognition of Angelicae Sinensis Radix.Methods:Stable isotope mass spectrometer(EA⁃IRMS)was used to determineδ^(13)C,δ^(15)N stable isotope ratios and percentage of carbon and nitrogen in 27 batches of Angelicae Sinensis Radix from three major production areas of Gansu(Mingui),Yunnan(Yungui)and Sichuan(Chuangui).The contents of seven trace elements(manganese,zinc,chromium,magnesium,iron,nickel,molybdenum)in the samples were determined by inductively coupled plasma mass spectrometer(ICP⁃MS).The stepwise linear discrimi⁃nant analysis(FLDA)was used for multivariate discriminant analysis.Results:The stable isotope ratios ofδ^(13)C,δ^(15)N in Angelicae Sinen⁃sis Radix from different producing areas showed differences.The contents of magnesium,chromium and iron in Angelicae Sinensis Radix were relatively rich.The content of molybdenum in Angelicae Sinensis Radix from Sichuan was relatively high,the contents of manga⁃nese,magnesium,iron,nickel and zinc in Angelicae Sinensis Radix from Yunnan were rich.FLDA with stable isotopes and the percent⁃age of carbon and nitrogen as discriminant variables had been tested by backward generation and cross⁃validation,the accuracy of origin discrimination was up to 90%and 83%,the prediction accuracy of external samples was 63%,the differentiation degree of large produ⁃cing areas(provincial administrative regions)was good,while that of small producing areas(county and municipal administrative re⁃gions)was not high.After the addition of trace elements as discriminant variables,the FLDA back⁃replacement accuracy reached 100%,the cross validation accuracy reached 91%and the prediction accuracy of external samples reached 100%.Conclusion:Theδ^(13)C,δ^(15)N stable isotopes,the percentage of carbon and nitrogen and trace eleme
