摘要
目的:建立墨旱莲HPLC指纹图谱研究方法,结合化学计量法对不同产地和不同干燥方法的墨旱莲质量进行比较。方法:采用Venusil XBP C_(18)(L)(250mm×4.6mm,5μm)色谱柱,流动相为0.1%磷酸溶液⁃甲醇,梯度洗脱;检测波长为263 nm;流速为1.0mL/min;柱温为25℃;建立不同产地和不同干燥方法的墨旱莲指纹图谱,利用相似度评价、聚类分析、主成分分析和偏最小二乘判别分析等化学计量方法对墨旱莲进行质量评价。结果:建立了墨旱莲HPLC指纹图谱,共确定30个共有峰,指认4个成分。不同产地墨旱莲样品可分为2类;晒干和50℃烘干的墨旱莲样品整体质量较好。结论:该指纹图谱研究方法简便可靠,重复性良好,通过指纹图谱结合化学计量分析的方法,明确了不同产地和不同干燥方法墨旱莲之间的差异,为墨旱莲及其制剂的内在质量评价提供参考。
Objective:To establish a HPLC fingerprint method of Ecliptae Herba and compare the quality of Ecliptae Herba from different habitats and different drying methods by chemometrics methods.Methods:The Venusil XBP C_(18)(L)(250 mm×4.6 mm,5μm)chromatographic column was used,the mobile phase was 0.1%phosphoric acid solution⁃acetonitrile with gradient elution.The detection wavelength was 263 nm,flow rate was 1.0 mL/min,column temperature was 25℃;The fingerprints of Ecliptae Herba with different producing areas and different drying methods were established,chemometrics methods such as similarity evaluation,cluster analysis,principal component analysis and partial least square discriminant analysis were used to evaluate the quality of Ecliptae Herba.Results:The HPLC fingerprints of Ecliptae Herba were established,a total of 30 common peaks were determined.The Ecliptae Herba samples from different producing areas could be divided into two categories.The overall quality of samples dried in sunshine and dried at 50℃was good.Conclusion:The fingerprint research method is simple and reliable,with good repeatability.By combining fingerprint analysis with chemometrics analysis,the differences of Ecliptae Herba between different producing areas and different drying methods are identified,which can provide reference for the internal quality evaluation of Ecliptae Herba and its preparations.
