摘要
Wnt/β-catenint信号通路广泛参与听觉器官发育、听觉细胞的增殖和分化,其异常调控导致了遗传性、老年性、药物性耳聋等听力疾病的发生或进展。胚胎发育过程中Wnt/β-catenint信号通路的异常表达会导致听觉器官形态和功能上的异常,β-catenin的缺失会抑制下游基因的转录,减少抗氧化因子的表达,提高听觉细胞的敏感性,引起毛细胞、听神经细胞抗损伤能力的下降。相反的,Wnt/β-catenin信号通路的激活能够抑制细胞的凋亡,减少毛细胞的损伤,甚至促进毛细胞增殖再生,Wnt/β-catenin信号通路为听力疾病的治疗提供了新的靶点。文章总结了Wnt/β-catenin信号通路调控听觉系统的最新研究进展,为听力疾病的分子生物学机制研究与临床治疗提供理论基础。
The Wnt/β-catenint signaling pathway is widely involved in the development of auditory organs,prolif-eration and differentiation of auditory cells.Abnormal regulation by the Wnt/β-catenin signaling pathway leads to the oc-currence or progression of genetic,age-related and drug-induced deafness and other hearing diseases.Dysregulation in expression of the Wnt/β-catenint signaling pathway during embryonic development leads to morphological and function-al abnormalities in auditory organs.Absence ofβ-catenin inhibits transcription of downstream genes,reduces the expres-sion of antioxidant molecules,increases the sensitivity of auditory cells,and causes damage to hair cells and nerve cells,leading to decreased hearing.Conversely,activation of the Wnt/β-catenin signaling pathway can inhibit cell apoptosis,reduce hair cell damage and promote hair cell proliferation and regeneration,which provides a new target for the treat-ment of hearing diseases.This paper summarizes the latest research progress on the Wnt/β-catenin signaling pathway in regulating hearing diseases,and provides a theoretical basis for molecular biological mechanisms and treatment of hear-ing diseases.
