摘要
目的观察外用小檗碱乳膏对耳廓复合痤疮模型大鼠皮损及血清中胰岛素样生长因子-1(insulin-like growth factor 1,IGF-1)、脱氢表雄酮(dehydroepiandrosterone,DHEA)因子的影响。方法将54只SPF级Wistar大鼠随机分为空白组、模型组、对照组、小檗碱乳膏高、中、低剂量组,共6组,每组9只。除空白组外,其余5组均建立大鼠耳廓复合痤疮模型,造模21 d。造模结束后,空白组及模型组的大鼠右耳廓处外用空白乳膏基质,对照组外用甲硝唑凝胶,小檗碱乳膏低、中、高剂量组分别外用0.011 g/(cm^(2)·d)、0.022 g/(cm^(2)·d)、0.044 g/(cm^(2)·d)剂量的小檗碱乳膏,涂抹给药,治疗28 d。肉眼观察各组大鼠皮损变化及镜下观察组织病理学变化情况,酶联免疫吸附试验(ELISA)测大鼠血清中IGF-1、DHEA因子的含量,采用单因素方差分析(one-way ANOVA)进行统计学分析。结果治疗28 d后,外用小檗碱乳膏对于耳廓复合痤疮模型大鼠皮损的红肿、脓疱、脱屑及病理情况作用显示,高、中、低剂量组及对照组与模型组比较差异均有统计学意义(P<0.05)。对于模型组大鼠血清中IGF-1、DHEA含量的调节作用,高、中、低剂量组及对照组均有降低(P<0.05),其中,中剂量组与对照组相比差异具有统计学意义(P<0.05)。结论外用小檗碱乳膏可显著改善大鼠耳廓复合痤疮模型的皮损,可能与其能够降低大鼠血清中IGF-1、DHEA含量,调节痤疮大鼠代谢相关。
Objective To observe the effects of external berberine cream on insulin-like growth factor 1(IGF-1)and Dehydroepiandrosterone(DHEA)in skin lesions and serum of rats with auricle combined acne.Methods Fifty-four SPF Wistar rats were randomly divided into six groups:blank group,model group,control group,high-dose,medium-dose and low-dose berberine cream groups,9 rats in each group.Except for blank group,the other five groups made auricle acne complex model for 21 days.After modeling,blank cream matrix was applied to the right ear of rats in the blank group and model group,metronidazole gel was applied to the control group,and 0.011 g/(cm^(2)·d),0.022 g/(cm^(2)·d)and 0.044 g/(cm^(2)·d)cream were applied to low-dose,medium-dose and high-dose berberine cream groups,respectively,for 28 days.The changes of skin lesions in each group were observed by naked eyes and the histopathological changes were observed under microscope.The contents of IGF-1 and DHEA in serum of the rats were measured by enzym-linked immunosorbent assay(ELISA).One-way ANOVA was used for statistical analysis.Results After 28 days of treatment,the effect of berberine cream on the redness,pustules,desquamate and pathological conditions of auricle compound acne model rats was compared,and the high,medium and low dose groups and the control group were significantly improved compared with the model group(P<0.05).The regulation effect of serum IGF-1 and DHEA contents in model group was significantly decreased in high,medium and low dose groups and control group(P<0.05),among which,the results of medium dose group were statistically different from control group(P<0.05).Conclusion Topical application of berberine cream can significantly improve the skin lesions of auricle in acne model rats,which may be related to the decrease of IGF-1 and DHEA contents in serum of rats and the regulation of metabolism of acne rats.
