摘要
研究香草乙酮对17型胶原蛋白(COL17A1)基因表达的“量效关系”和安全剂量,在此基础上开发新型抗衰老护肤产品并检验实际功效。体外培养HaCaT细胞(人永生化角质形成细胞株),用不同浓度的香草乙酮处理细胞,实时定量PCR法(RT-PCR)检测HaCaT细胞中COL17A1基因mRNA的转录水平,Western blot检测COL17A1蛋白表达水平;利用鸡胚绒毛膜尿囊膜血管毒性实验(CAMVA)确定既安全又有效诱导COL17A1表达的香草乙酮浓度制备护肤品,并在28名中老年志愿者皮肤上进行功效测实。结果表明,经香草乙酮处理后,HaCaT细胞中COL17A1基因表达量在一定浓度范围内显著提高,当香草乙酮浓度达到60μmol/L时,COL17A1基因表达量最高;浓度为120μmol/L时,COL17A1基因表达量开始呈下降趋势。香草乙酮浓度小于8400μmol/L时具有良好安全性;人体皮肤皱纹严重程度在使用香草乙酮后得到一定缓解。
Apocynin has been proved to induce the gene expression of COL17A1 collagen.To provide a direction for the development of new anti-aging products,the effects of apocynin on gene expression of COL17A1 collagen in HaCaT cells were studied and its efficacy on skin was tested.HaCaT cells were cultured in vitro and treated with different concentrations of apocynin.Then the transcription levels of COL17A1 mRNAs were measured by RT-PCR.The expression of COL17A1 was tested by Western blot.The safety of a product which was prepared with appropriate concentration of apocynin,was determined by CAMVA,and skin experiments were conducted on 28 middle-and old-aged volunteers.When HaCaT cells were treated with apocynin,the gene expression of COL17A1 was significantly increased in a certain concentration range.The gene expression reached the highest level when the concentration of apocynin was 60μmol/L,and began to decrease when the concentration of apocynin reached 120μmol/L.The results of CAMVA experiment showed that the products of apocynin had no irritation to human skin.Human skin test showed that the product had a certain therapeutic effect on human skin aging.
