基于基因芯片检测技术探讨NF-κB信号通路在HIV/AIDS肺脾气虚证中的表达和作用 被引量:5

Discussion on expression and role of NF-κB signaling pathway in HIV/AIDS patients with lung and spleen qi deficiency syndrome based on gene chip detection technology

摘要 目的:通过对艾滋病(HIV/AIDS)肺脾气虚证患者NF-κB信号通路相关基因检测分析,探讨NF-κB信号通路在HIV/AIDS肺脾气虚证中的作用机制。方法:采用定制的RT2 Profiler PCR Array芯片检测HIV/AIDS肺脾气虚证证患者外周血单个核细胞中NF-κB信号通路相关基因表达情况。结果:依据按照2-ΔΔC t值>1.5或2-ΔΔCt值<0.67、P<0.05标准筛选发现,HIV/AIDS肺脾气虚证组与健康对照组对比较有12个差异表达基因,其中表达量降低的基因有8个(IL-1B、IL-1R1、TLR4、TRAF2、NF-κB1、BCL10、CFLAR、PTGS2),表达量升高的基因有4个(BCL2A1、CCL4、TNFAIP3、NFκBIA)。结论:NF-κB信号通路对HIV/AIDS肺脾气虚证在凋亡、免疫、炎症等方面具有调节作用,差异表达基因可能是调控HIV/AIDS肺脾气虚证的部分机制。 Objective:Through the detection and analysis of NF-κB signaling pathway related genes in HIV/AIDS patients with lung and spleen qi deficiency syndrome,to explore the mechanism of NF-κB signaling pathway in HIV/AIDS lung and spleen qi deficiency syndrome.Methods:Customized RT2 Profiler PCR Array chips was used to detect the expression of genes related to NF-κB signaling pathway in peripheral blood mononuclear cell of HIV/AIDS patients with lung and spleen qi deficiency syndrome.Results:According to the criteria of flod Chang>1.5 or flod Chang<0.67,P<0.05,it was found that there were 12 differentially expressed mRNA in the HIV/AIDS lung and spleen qi deficiency syndrome group compared with the healthy control group,among which 8 genes with reduced expression were IL-1B,IL-1R1,TLR4,TRAF2,NF-κB1,BCL10,CFLAR,PTGS2,and 4 genes with increased expression were BCL2A1,CCL4,TNFAIP3,NF-κBIA.Conclusion:NF-κB signaling pathway has regulatory effects on apoptosis,immunity,inflammation and other aspects of HIV/AIDS lung and spleen qi deficiency syndrome.Differentially expressed genes may be part of the mechanism regulating HIV/AIDS lung and spleen qi deficiency syndrome.
