摘要
目的建立快速检测血样中氟马西尼、咪达唑仑、罗哌卡因和地佐辛浓度的方法。方法血样经乙腈沉淀蛋白后,用HPLC-MS/MS检测,在ESI正离子模式下结合多离子反应检测模式,对待测血样进行分析测定。结果待测药物在一定线性范围内,线性关系良好,回收率在95.6%~109.8%,日内精密度在1.06%~4.48%,日间精密度在2.23%~7.48%,氟马西尼最低检出限为0.5 ng/ml,定量限为2 ng/ml;罗哌卡因检出限为0.3 ng/ml,定量限为1 ng/ml;地佐辛检出限为1 ng/ml,定量限为3 ng/ml;咪达唑仑为0.5 ng/ml,定量限为2 ng/ml。结论本文建立了快速检测血中4种麻醉药物的方法,为临床血药浓度监测和司法鉴定提供了依据。
Objective To establish a rapid method for the determination of flumazenic,midazolam,ropivacaine and dezocine in blood samples.Methods Blood samples were treated with acetonitrile protein precipitation,then detected by HPLC-MS/MS under ESI positive ion mode combined with multi-ion reaction detection mode.Results Good linearity was obtained for the four anesthetic drugs,with the recovery ranging from 95.6%to 109.8%,the intra-day precision ranging from 1.06%to 4.48%,and the inter-day precision ranging from 2.23%to 7.48%.The limit of detection for flumazenil was 0.5 ng/ml,and the limit of quantitative was 2 ng/ml.The limit of detection for ropivacaine was 0.3 ng/ml,and the limit of quantitative was 1 ng/ml.The limit of detection for dezocine was 1 ng/ml,and the limit of quantitative was 3 ng/ml.The limit of detection for midazolam was 0.5 ng/ml,and the limit of quantitative was 2 ng/ml.Conclusion The rapid detection method for the four anesthetic drugs in blood was established,which can be applied in drug monitoring and forensic science.
