摘要
目的探讨电针神阙穴对功能性便秘水通道蛋白3(aquaporins 3,AQP3)和核转录因子-κB(nuclear factor-κB,NF-κB)信号通路及胃肠动力的影响。方法将40只健康SD大鼠随机分为空白组、模型组、苁蓉通便口服液组、电针治疗组,每组10只。模型复制成功后,按相应组别处理方法连续干预14 d。测定大鼠粪便粒数、粪便含水量及炭末推进率,苏木精-伊红染色观察结肠组织病理变化,ELISA检测结肠组织胃动素(motilin,MTL)、胃泌素(gastrin,GAS)、P物质(substance P,SP)、环氧化酶-2(cyclooxygenase-2,COX-2)、白细胞介素-1β(interleukin-1β,IL-1β)、白细胞介素-6(interleukin-6,IL-6)、肿瘤坏死因子-α(tumor necrosis factor-alpha,TNF-α)含量,qRT-PCR检测结肠组织AQP3、NF-κB p65 mRNA表达水平,Western blot检测结肠组织AQP3、囊性纤维化跨膜传导调节因子(cystic fibrosis transmembrane conductance regulator,CFTR)、蛋白激酶A(protein kinase A,PKA)、磷酸化核转录因子-κB p65(phosphorylated nuclear transcription factor-κB p65,p-NF-κB p65)、核因子κB抑制蛋白α(inhibitor of nuclear factor-kappa B,IκBα)表达水平。结果与空白组比较,模型组大鼠粪便粒数、粪便含水量及炭末推进率明显减少(P<0.05),结肠组织黏膜层坏死较为明显,结肠组织MTL、GAS、SP含量及AQP3 mRNA和蛋白表达水平,CFTR、PKA、IκBα蛋白表达水平明显降低(P<0.05),COX-2、IL-1β、IL-6、TNF-α含量,NF-κB p65 mRNA表达水平及p-NF-κB p65蛋白表达水平明显升高(P<0.05);与模型组比较,电针治疗组的粪便粒数,粪便含水量,炭末推进率,MTL、GAS、SP含量明显升高(P<0.05),结肠组织COX-2、IL-1β、IL-6、TNF-α含量及NF-κB p65 mRNA表达水平明显降低(P<0.05),结肠组织AQP3 mRNA和蛋白表达水平,CFTR、PKA、IκBα蛋白表达水平明显升高(P<0.05)。结论电针神阙穴能够改善功能性便秘大鼠粪便含水量和肠道转运速率,其机制可能与调节AQP3和NF-κB信号通路表达有�
Objective To investigate the effect of acupuncture at Shenque point on the aquaporin-3(AQP3)and nuclear factor-kappa B(NF-κB)signaling pathways and gastrointestinal motility in functional constipation.Methods A total of 40 healthy Sprague-Dawley rats were randomly divided into blank group,model group,Congrong Tongbian Oral Liquid group,and acupuncture treatment group,with 10 rats in each group.After successful modeling,the rats were given corresponding treatment for 14 consecutive days.Number of fecal pellets,fecal water content,and charcoal propulsion rate were measured;HE staining was used to observe the pathological changes of colon tissue;ELISA was used to measure the levels of motilin(MTL),gastrin(GAS),substance P(SP),cyclooxygenase-2(COX-2),interleukin-1β(IL-1β),interleukin-6(IL-6),and tumor necrosis factor-alpha(TNF-α)in colon tissue;qRT-PCR was used to measure the mRNA expression of AQP3 and NF-κB p65 in colon tissue;Western blot was used to measure the protein expression of AQP3,cystic fibrosis transmembrane conductance regulator(CFTR),protein kinase A(PKA),NF-κB p65,and inhibitor of nuclear factor-kappa B(IκBα)in colon tissue.Results Compared with the blank group,the model group had significant reductions in the number of fecal pellets,fecal water content,and charcoal propulsion rate(P<0.05),obvious mucosal necrosis of colon tissue,and significant reductions in the levels of MTL,GAS,and SP,the mRNA and protein expression levels of AQP3,and the protein expression levels of CFTR,PKA,and IκBαin colon tissue(P<0.05),as well as significant increases in the levels of COX-2,IL-1β,IL-6,and TNF-α,the mRNA expression level of NF-κB p65,and the protein expression level of p-NF-κB p65(P<0.05).Compared with the model group,the acupuncture treatment group had significant increases in the number of fecal pellets,fecal water content,charcoal propulsion rate,and the levels of MTL,GAS,and SP(P<0.05),significant reductions in the levels of COX-2,IL-1β,IL-6,and TNF-αand the mRNA expression level of NF-κB p
