S100A6通过抑制细胞分裂、迁移、侵袭及促进凋亡抑制Calu-6肺癌细胞株生长 被引量:9

S100A6 represses Calu-6 lung cancer cells growth via inhibiting cell proliferation,migration,invasion and enhancing apoptosis

摘要 目的探究S100A6对肺癌细胞株Calu-6增殖、迁移、凋亡等的生物学行为影响。方法实验性研究。构建S100A6过表达慢病毒载体,和空载体同时转染至Calu-6肺癌细胞株。转染成功后(RT-PCR、Western-blot确认S100A6过表达),使用MTT、Transwell和流式细胞术实验技术分别检测Calu-6/S100A6实验组(Calu-6/S100A6)、Calu-6/空载体对照组(Calu-6/neo)、Calu-6空细胞对照组(Calu-6)的细胞增殖能力、细胞迁移和侵袭能力、细胞凋亡和周期,比较3组差异。将3组细胞注入裸鼠皮下(15只),每周测定小鼠成瘤体积。结果Calu-6肺癌细胞株转染S100A6后与Calu-6/neo、Calu-6组比较,Calu-6/S100A6组的增殖、迁移和侵袭能力均呈减弱趋势,细胞凋亡能力增强,细胞周期G1期延长,S期缩短(P<0.05)。实验5周后测定Calu-6/S100A6组肿瘤体积明显小于Calu-6/neo和Calu-6组(P值均<0.05)。结论S100A6抑制Calu-6肺癌细胞的生长,同时减弱Calu-6细胞株的成瘤能力。在细胞水平上,S100A6有抑癌功能。 Objective To explore the biological effects of S100A6 on proliferation,migration and apoptosis of lung cancer cell line Calu-6.Methods This was an experimental research.The S100A6 overexpression lentiviral vector was constructed and transfected into Calu-6 lung cancer cell line simultaneously with the empty vector.After successful transfection(S100A6 overexpression was confirmed by RT-PCR,Western blot),MTT,Transwell and flow cytometry were used to detect the cell proliferation,migration and invasion,apoptosis and cycle of Calu-6/S100A6 experimental(Calu-6/S100A6)group,Calu-6/empty vector(Calu-6/neo)control group and Calu-6 empty cell(Calu-6)control group,respectively.The three groups of cells were injected subcutaneously into nude mice(15 mice),and the tumor volume of mice was measured every week.Results Compared with Calu-6/neo and Calu-6 group,Calu-6/S100A6 group showed a weakening trend in the cell behaviors of proliferation,migration and invasiveness,while had an enhancement of cell apoptosis(all P<0.05).The cell cycle of Calu-6/S100A6 cells had an increase of G1 phase but a reduction of S phase(P<0.05).After five weeks of cell injection,the tumor bulk of Calu-6/S100A6 group was smaller than that of Calu-6/neo or Calu-6 group(both P<0.05).Conclusions S100A6 inhibits the growth of Calu-6 lung cancer cells and impairs the ability in tumorigenesis of Calu-6.At cellular level,S100A6 is supposed to act as a tumor suppressor gene in lung cancer.
