玫瑰黄酮提取物对高脂血症大鼠血脂水平和抗氧化作用的研究 被引量:2

摘要 目的研究玫瑰黄酮提取物(RFE)对高脂血症大鼠血脂的影响及其抗氧化作用。方法SD大鼠60只,随机分为对照组、模型组、阳性药物组及玫瑰黄酮提取物低剂量组(RFE-L组,6 mg/kg)、中剂量组(RFE-M组,12 mg/kg)、高剂量组(RFE-H组,24 mg/kg),每组10只。对照组给予普通饲料喂养,其余组均以高脂饲料喂养8 w诱发大鼠高脂血症模型。成模后药物干预:阳性药物组给予4 mg/kg体质量辛伐他汀灌胃,玫瑰黄酮提取物按低、中、高剂量灌胃,模型组和对照组给予同等体积生理盐水灌胃,每日1次,连续1个月。灌胃结束取大鼠全血及肝脏,用于检测血脂水平、谷胱甘肽(GSH-Px)、丙二醛(MDA)、超氧化物歧化酶(SOD)及肝脏病理改变。结果经高脂饲料饲养8 w后,检测血脂水平,高脂血症动物成模率为76%。各组大鼠的肝脏病理改变:模型组动物肝脏肝小叶结构被破坏,可见大量脂肪空泡,RFE-L、RFE-M、RFE-H组随着药物浓度的增加,肝脏病变逐渐减轻。药物干预后大鼠血脂水平:与对照组比较,模型组TC、TG、LDL-C显著升高,HDL-C显著降低,差异有统计学意义(P<0.01)。与模型组比较,阳性药物组、RFE-L、RFE-M、RFE-H组中大鼠血清中TC、TG、LDL-C水平降低,HDL-C水平升高,差异有统计学意义(P<0.05)。抗氧化指标检测结果显示在不同组SOD、MDA、GSH-Px的改变不同,差异有统计学意义(P<0.01)。SOD和GSH-Px随着药物浓度升高而升高,MDA随着药物浓度升高而降低,与模型组比较,阳性药物、RFE-M、RFE-H组差异均有统计学意义(P<0.05);RFE-M、RFE-H组SOD和GSH-Px随着药物浓度升高而升高,MDA随着药物浓度升高而降低,与RFE-L组比较,RFE-M、RFE-H组差异均有统计学意义(P<0.05)。结论玫瑰黄酮提取物的降血脂作用可能与机体的抗氧化酶活性有关,且与玫瑰黄酮提取物呈剂量依赖性。 Objective To study the effect of rose flavone extract(RFE)on the liver of hyperlipidemia rats and its antioxidant effect.Methods A total of 60 SD rats were randomly divided into control group,model group,positive drug(simvastatin 4 mg/kg body weight)group and rose flavone extract low-,medium-and high-dose group(RFE-L,M and H:6,12 and 24 mg/kg),with 10 rats in each group.Except the control group,the other groups were fed with high-fat feed for 8 weeks to induce hyperlipidemia model in rats and then were intervented with drugs by gavage once a day in one month.Whole blood and liver of rats were taken to detect blood lipid levels,glutathione(GSH-Px),malondialdehyde(MDA),superoxide dismutase(SOD)and liver pathological changes.Results After being fed with high-fat feed for 12 weeks,the blood lipid level was tested,and the model rate of hyperlipidemia animals was 76%.Liver hyperlipidemic pathological changed of rats in each group were observed.The liver disease gradually reduced with dose dependent of RFE.Compared with the control group,TC,TG,and LDL-C in the model group were significantly increased,and HDL-C was significantly decreased(all P<0.01).Compared with the model group,the serum levels of TC,TG and LDL-C in the positive drug group,RFE-L,M,and H groups decreased,and the HDL-C level increased(all P<0.05).Antioxidant index test results showed that the changes of SOD,MDA,GSH-Px in different groups were different(P<0.01).Compared with the model group,SOD and GSH-Px increased,and MDA decreased with dose-dependence,and the difference between the positive drug group,and RFE-M,-H is statistically significant(P<0.05);compared with the RFE-L,-M,-H group,SOD and GSH-Px increased and MDA decreased with dose-dependence(P<0.05).Conclusion The hypolipidemic effect of rose flavone extract may be related to the body′s antioxidant enzyme activity dose-dependently.
