摘要
目的探讨犬急性冠脉闭塞后血小板-白细胞聚集体(PLA)的变化特点。方法选取10~13个月龄的犬15只,抽取犬静脉血体外制备混合性血栓,经介入术将体外制备的混合性血栓置入冠状动脉内建立犬急性冠脉闭塞模型。于术前、闭塞时、闭塞后30 min、1 h、2 h采集比格犬新鲜静脉血,用流式细胞仪测定血小板α-颗粒膜糖蛋白(CD62P)阳性率及血小板-白细胞结合物(PLA)、血小板-单核细胞聚集体(PMA)、血小板-中性粒细胞聚集体(PNA)、血小板-淋巴细胞聚集体(PlyA)占总白细胞T、单核细胞、粒细胞、淋巴细胞的百分率(PLA%、PMA%、PNA%、PlyA%),进行分析。于术前、闭塞后2 h、4 h采集比格犬新鲜动脉血,测定犬血清中肌钙蛋白T(cTnT)的含量。处死动物,心肌组织做病理HE染色检查。结果与术前比较,CD62P阳性率于闭塞时、闭塞后30 min、1 h、2 h均降低(P<0.05),于闭塞后2 h降低最明显(P<0.05);PLA%于闭塞时、闭塞后30 min、1 h、2 h均增高(P<0.05);PMA%于闭塞后2 h增高(P<0.05);PNA%于闭塞后30 min、1 h、2 h均增高(P<0.05);cTnT于闭塞后2 h未见明显变化,闭塞后4 h增高(P<0.05)。HE染色结果示心肌纤维排列整齐,未见心肌灶状坏死及炎细胞浸润。结论犬处于急性冠脉闭塞时,于闭塞后30 min、1 h、2 h,PLA水平显著增高,且提前于cTnT出现变化。
Objective To investigate the changes of platelet leukocyte aggregates(PLA)after acute coronary occlusion in dogs.Methods The mixed coronary artery occlusion model was established in beagle dogs aged from 10 months to 15 months.Fresh venous blood was collected from the dogs before operation,at the time of occlusion,30min,1h and 2h after occlusion.The positive rate of plateletα-granμLe membrane glycoprotein(CD62P),platelet leukocyte conjugate(PLA),platelet monocyte aggregation(PMA),and platelet intermediate granμLocyte aggregation(PNA)were measured by flow cytometry,The percentage of PLYA in total leukocytes,monocytes,granμLocytes and lymphocytes(PLA%,PMA%,PNA%,PLYA%)were analyzed.Fresh arterial blood was collected from beagle dogs before operation,2h and 4h after occlusion,and the content of serum troponin T(cTnT)was determined.And the myocardial tissues were examined by HE staining.Results Compared with the preoperative,CD62P positive rate decreased at occlusion time,30min,1 h and 2h after occlusion(P<0.05),and decreased most significantly at 2 h after occlusion(P<0.05);PLA%increased at 30min,1h,2h after occlusion(P<0.05);PMA%increased at 2h after occlusion(P<0.05);PNA%increased at 30min,1h,2h after occlusion(P<0.05);cTnT did not change significantly at 2h after occlusion,then increased at 4h after occlusion(P<0.05).HE staining showed that the myocardial fibers were arranged in order,and there was no myocardial focal necrosis and inflammatory cell infiltration.Conclusion When the dog was in acute coronary occlusion,HE staining resμLts showed that there was no myocardial focal necrosis and inflammatory cell infiltration.At 30min,1h,2h after occlusion,PLA level was significantly increased,and cTnT began to increase at 4h after occlusion.
