蒙脱石对黄曲霉毒素B_(1)体外吸附效果评估

摘要 试验评估蒙脱石对黄曲霉毒素B_(1)(AFB_(1))的体外吸附效果,探究AFB_(1)浓度、蒙脱石有效含量及添加量、pH值、吸附时间及吸附温度对蒙脱石吸附AFB_(1)的影响。试验制作高(98.8μg/kg)、低(48.9μg/kg)两种不同浓度的AFB_(1)污染饲料,将5种不同有效含量的蒙脱石以3种添加量添加到带毒饲料中吸附AFB_(1),加入提取液后离心取上清液,采用ELISA法测定上清液中残余AFB_(1)含量,并计算吸附率;确定吸附效果较好的蒙脱石有效含量及添加量后,在不同pH值(2~7)、吸附时间(5~115 min)及吸附温度(20~40℃)条件下测定蒙脱石对AFB_(1)的吸附率。结果表明,高毒素浓度下蒙脱石吸附效果高于低毒素浓度下蒙脱石吸附效果。在蒙脱石有效含量为70%~92%范围内,AFB_(1)的吸附率与蒙脱石有效含量呈正相关。只有蒙脱石有效含量高于90%时,对AFB_(1)才达到80%以上的良好吸附效果,含量高于95%后,蒙脱石对AFB_(1)的吸附效果较为稳定;不同蒙脱石添加量的吸附效果为0.4%添加量>0.3%添加量>0.2%添加量。pH值对蒙脱石脱毒效果影响不明显,pH值在2~7范围内,AFB_(1)吸附率均在90%以上。蒙脱石对AFB_(1)的吸附在55 min时接近平衡,此时吸附率为93.2%;55 min后吸附曲线趋近平稳;95 min时吸附率最高,为94.8%。在20~40℃范围内,温度越高,吸附效果越好;35℃时,吸附率最高,为93.2%。 The experiment was to evaluate the adsorption effect of montmorillonite on AFB_(1)in vitro,and to explore the effect of AFB_(1)concentration,effective content and addition amount of montmorillonite,pH value,adsorption time and adsorption temperature on adsorption of AFB_(1)by montmorillonite.Two kinds of AFB_(1)contaminated feeds with high(98.8μg/kg)and low(48.9μg/kg)concentrations were prepared.Five kinds of montmorillonite with different effective contents were added into contaminated feed with three kinds of supplemental amounts to adsorb AFB_(1),and the supernatant was centrifugally extracted after adding the extract.The residual AFB_(1)content in supernatant was determined by ELISA,and the adsorption rate was calculated.After determining the effective content and addition amount of montmorillonite with better adsorption effect,the adsorption rate of montmorillonite on AFB_(1)was determined under different pH value(2~7),adsorption time(5~115 min)and adsorption temperature(20~40℃).The results showed that the adsorption effect of montmorillonite at high toxin concentration was higher than that at low toxin concentration.When the effective content of montmorillonite was 70%~92%,the adsorption rate of AFB_(1)was positively correlated with the effective content of montmorillonite.Only when the effective content of montmorillonite was higher than 90%,the good adsorption effect of AFB_(1)can reach more than 80%.When the effective content of montmorillonite was higher than 95%,the adsorption effect of montmorillonite on AFB_(1)was relatively stable.The adsorption effect of different adding amount of montmorillonite was 0.4%adding amount>0.3%adding amount>0.2%adding amount.The effect of pH value on detoxification effect of montmorillonite was not obvious.The adsorption rate of AFB_(1)was above 90%in the range of pH 2~7.The adsorption of montmorillonite on AFB_(1)was close to equilibrium at 55 min,and the adsorption rate reached 93.2%.After 55 min,the adsorption curve was nearly stable,and the adsorption rate reach
