酵母硒对滩羊的生物安全性评价:生长性能、血液常规参数、硒蛋白基因表达以及富集规律 被引量:8

摘要 本试验旨在研究饲粮酵母硒添加水平对滩羊生长性能、血液常规参数、硒蛋白基因表达以及组织器官硒含量的影响,进而评价酵母硒对滩羊的生物安全性,揭示硒在滩羊体内的生物富集规律。选取体重[(32±2)kg]相近、健康状况良好的滩羊公羔64只,随机分为4组,每组16个重复,每个重复1只羊。试验选用玉米-豆粕型基础饲粮,硒含量为0.16 mg/kg,各组分别在基础饲粮中添加28.94、105.46、258.51、564.60 mg/kg酵母硒,使饲粮硒含量分别为0.25(Ⅰ组)、0.50(Ⅱ组)、1.00(Ⅲ组)、2.00 mg/kg(Ⅳ组)。预试期10 d,正试期60 d。结果表明:1)饲粮酵母硒添加水平对滩羊生长性能无显著影响(P>0.05)。2)与Ⅰ组相比,Ⅲ组滩羊血液平均红细胞体积、嗜酸性粒细胞计数显著升高(P<0.05)。与Ⅱ组相比,Ⅳ组滩羊血液红细胞分布宽度标准差显著降低(P<0.05)。3)与Ⅰ组相比,Ⅲ组滩羊肝脏谷胱甘肽过氧化物酶1(GPX1)、谷胱甘肽过氧化物酶3(GPX3)的mRNA相对表达量呈二次极显著升高(P<0.01)。与Ⅰ组相比,Ⅱ、Ⅲ、Ⅳ组滩羊肝脏硫氧还原白还原酶1(TXNRD1)的mRNA相对表达量呈二次极显著下降(P<0.01),甲状腺激素脱碘酶1(DIO1)的mRNA相对表达量呈线性显著下降(P<0.05)。各组之间滩羊肌肉硒蛋白mRNA相对表达量均无显著差异(P>0.05)。4)Ⅳ组滩羊血清硒含量极显著高于Ⅰ、Ⅱ、Ⅲ组(P<0.01)。随着饲粮酵母硒添加水平的提高,滩羊背肌、肝脏、肾脏、肺脏、心脏、胰腺、十二指肠硒含量呈二次极显著增加(P<0.01),脾脏、睾丸硒含量呈线性极显著增加(P<0.01)。综上所述,以酵母硒为补充硒源,饲粮硒含量在0.25~2.00 mg/kg时对滩羊生长性能、血液常规参数、硒蛋白基因表达以及组织器官硒含量均无不良影响;饲粮硒含量达2.00 mg/kg时饲喂滩羊是安全的。滩羊血清及组织器官硒富集量随着饲粮酵母硒添加水平的提高而增加。 This study was conducted to evaluate the effects of dietary selenium yeast supplemental level on growth performance,blood routine parameters,selenoprotein gene expression and tissue and organ selenium contents of Tan sheep,and to evaluate the biological safety of selenium yeast for Tan sheep and reveal the biological enrichment regularity of selenium.Sixty-four healthy male Tan sheep with similar body weight[(32±2)kg]were selected and randomly assigned into 4 groups with 16 replicates in each group and 1 sheep in each replicate.The content of selenium in corn-soybean meal basal diet was 0.16 mg/kg,and 28.94,105.46,258.51 and 564.60 mg/kg selenium yeast were added to the basal diet,to make the contents of selenium in diets were 0.25(groupⅠ),0.50(groupⅡ),1.00(groupⅢ)and 2.00(groupⅣ)mg/kg,respectively.The pre-experimental period lasted for 10 days,and the experimental period lasted for 60 days.The results showed as follows:1)dietary selenium yeast supplemental level had no significant effects on growth performance of Tan sheep(P>0.05).2)Compared with groupⅠ,the mean corpuscular volume and eosinophil count in blood of Tan sheep of groupⅢwere significantly increased(P<0.05).Compared with groupⅡ,the standard deviation of red cell volume distribution width in blood of Tan sheep of groupⅣwas significantly decreased(P<0.05).3)Compared with groupⅠ,the mRNA relative expression levels of glutathione peroxidase 1(GPX1)and glutathione peroxidase 3(GPX3)in liver of Tan sheep of groupⅢwere significantly increased in a quadratic manner(P<0.01).Compared with groupⅠ,the mRNA relative expression level of thioredoxin reductase 1(TXNRD1)in liver of Tan sheep of groupsⅡ,ⅢandⅣwas significantly decreased in a quadratic manner(P<0.05),and the mRNA relative expression level of thyroxine deiodinase 1(DIO1)was significantly decreased in a linear manner(P<0.05).There were no significant differences in mRNA relative expression levels of selenoprotein in muscle among all groups(P>0.05).4)The serum selenium content o
