摘要
为研究早胜牛CD36基因第三外显子的多态性及其与生长及肉用性状的关联性,选取300头健康的早胜肉牛,采用PCR-SSCP技术、DNA测序技术对早胜牛CD36基因第三外显子进行遗传多态性分析,通过SPSS 19.0软件分析SNP位点与早胜牛生长性状和肉用性状的关联性,采用实时荧光定量PCR技术检测早胜牛不同组织器官中CD36基因的表达特性,分析其遗传变异与生长性能的关系。结果表明,CD36基因第三外显子在59415 bp处存在一个SNP,具有GG、GC和CC 3种基因型;关联分析显示,G59415C位点,CC和GC型体重极显著高于GG型(P<0.01),CC型体斜长显著高于GG型(P<0.05),CC型的宰前活重、胴体重显著高于GG型(P<0.05),背膘厚度显著高于GG型和GC型(P<0.05),肌内脂肪含量极显著高于GG和GC型(P<0.01);荧光定量PCR检测显示,腿肌、背最长肌和肝脏中mRNA表达量以CC型最高,GC型次之,GG型最低,差异不显著(P>0.05)。说明G59415C位点对早胜牛生长性状和肉用性状影响较大,可作为生长性状和肉用性状分子标记位点,CC型为优势基因型,可作为生长性状和肉用性状的候选基因型。
The aim of the study was to research polymorphism of the third exon of CD36 gene and its association with growth and meat traits of Zaosheng cattle,and to provide a theoretical basis for the improvement of Zaosheng cattle.300 healthy Zaosheng cattle were used to detect polymorphism of CD36 by means of DNA sequencing and PCR-SSCP.The association analysis was conducted between the CD36 gene SNP site and traits of growth and meat of Zaosheng cattle with SPSS 19.0 software.To analyze the relationship between genetic variation and growth performance,qPCR was used to detect the expression characteristics of CD36 gene in different tissues and organs.The results showed that the third exon of CD36 gene had a SNP at 59415 bp,with 3 genotypes(GG,GC and CC).The correlation analysis showed that at the G59415C site,the body weight of CC and GC were significantly higher than GG(P<0.01),the body oblique length of CC was significantly higher than GG(P<0.05),the live weight before slaughter and carcass weight of CC were significantly higher than GG(P<0.05),the dorsal standard thickness of CC was significantly higher than GG and GC(P<0.05),and the intramuscular fat content was significantly higher than GG and GC(P<0.01).qPCR showed that the mRNA expression levels of leg muscle,longest dorsal muscle and liver CC type were higher than GG and GC(P>0.05).The result showed that the G59415C locus has a great influence on growth traits and meat traits of Zaosheng cattle,and can be used as a molecular marker for growth traits and meat traits.CC type is the dominant genotype and can be used as a candidate genotype for growth traits and meat traits.
