摘要
目的探讨长链非编码RNA-00707(long-chain non-coding RNA-00707,Linc00707)在乳腺癌组织中定性与定量表达,及其与乳腺癌分子病理分型的相关性。方法建立人源脂肪间充质干细胞与人乳腺癌MCF7细胞间接共培养体系,采用LncRNAs芯片技术获得Linc00707,应用RNAscope技术检测136例乳腺癌及癌旁正常乳腺组织中Linc00707的表达;观察mRNA在癌细胞中的表达定位及按照mRNA表达量等级的判读结果,分析Linc00707表达与乳腺癌分子病理分型的相关性。结果136例乳腺癌组织中121例Linc00707阳性,呈细胞核及细胞质内点簇状棕色颗粒分布,乳腺癌及癌旁正常乳腺组织中Linc00707的阳性率分别为89%(121/136)、3.8%(3/78),Linc00707阳性定量评分1分占9.1%(11/121),2分占30.6%(37/121),3分占40.4%(49/121),4分占19.8%(24/121)。Luminal A型、Luminal B型、HER-2过表达型、基底样型乳腺癌中Linc00707的阳性率分别为52.6%(10/19)、98.1%(51/52)、94.9%(37/39)、88.5%(23/26)。Linc00707在乳腺癌组织中的阳性率高于癌旁正常乳腺组织,定量评分高评分(2、3、4分)组的阳性率高于低评分(1分)组,Luminal B型、HER-2过表达型、基底样型乳腺癌中的Linc00707阳性率高于Luminal A型乳腺癌。结论Linc00707可以促进乳腺癌的发生、发展,对乳腺癌的恶性程度评估有一定的临床意义及成为乳腺癌标志物具有潜在应用价值。
Purpose To study the qualitative and quantitative expression of long-chain non-coding RNA-00707(Linc00707)in breast cancer tissues,and to explore its correlation with molecular pathological classification of breast cancer.Methods An indirect co-culture system of human adipose-derived mesenchymal stem cells(HAD-MSCs)and human breast cancer MCF7 cells was established,and Linc00707 was obtained by lncRNAs chip technology.136 breast cancer tissue samples were collected,and the difference of expression of Linc00707 between cancer and adjacent cancer was detected by RNAscope technology.The location of mRNA expression in cancer cells was observed and the results were interpreted according to mRNA expression level,and the correlation between Linc00707 positive expression and pathological molecular subtypes of breast cancer was analyzed by Chi-square test.Results Linc00707 was positive in 121 cases of breast cancer,which was distributed in the nucleus and cytoplasm of cancer cells.The positive expression rates of Linc00707 in breast cancer tissues and adjacent to cancer were 89%(121/136)and 3.8%(3/78),respectively.The positive quantitative score of Linc00707 was 9.1%(11/121)in 1 score,30.6%(37/121)in 2 score,40.4%(49/121)in 3 score and 19.8%(24/121)in 4 score,the positive expression rates of luminal A type,Luminal B type,HER overexpression type and basal type were 52.6%(10/19),98.1%(51/52),94.9%(37/39)and 88.5%(23/26),respectively.The positive expression rate of Linc00707 in breast cancer tissue is higher than that in normal breast tissue adjacent to cancer.Quantitative score analysis shows that the expression rate of high score(2,3 and 4)is higher than that of low score(1),and the positive expression rates of luminal B type,HER overexpression type and basal type are higher than that of luminal A type.Conclusion Linc00707 can promote the occurrence and development of breast cancer.It has a certain clinical significance in evaluating the malignancy of breast cancer and has potential application value as a marker for breast
