摘要
目的:明确外感发热药浴方调控IL-6的主要活性成分并建立含量测定方法。方法:采用风寒刺激法制备大鼠外感风寒模型,以血清IL-6水平显著升高为造模成功;取模型动物血清,加入外感发热药浴方中活性成分单体溶液进行孵育,使用ELISA法测定IL-6水平,与模型组比较,选择明显下降者列为含量测定指标;含量测定使用高效液相色谱法,测定指标为麻黄碱和伪麻黄碱。结果:外感发热药浴方各成分中,白花前胡甲素、白花前胡乙素、麻黄碱、伪麻黄碱和连翘苷具有明显的IL-6调控活性,但仅有麻黄碱和伪麻黄碱在HPLC测定中有明显峰响应,二者含量分别为0.4215~0.4947mg/mL和0.1217~0.1915mg/mL。结论:麻黄碱和伪麻黄碱可作为评价外感发热药浴方调控IL-6的活性成分。
Objective:To clarify main IL-6 regulatory components in Waigan Fenghan medicated bath formula and establish simultaneous determination method.Methods:The rat cold model was constructed was wind and cold stimulation,the animal with obvious rise in serum IL-6 levels were regarded as successful model construction;serum of the model animal was taken and compound monomers of Waigan Fenghan medicated bath formula were added for incubation,the IL-6 levels of those groups were determined by ELISA,components with significant decrease compared with model were chosen as indexes for content determination;and the latter was carried out by high performance liquid chromatography,with ephedrine and pseudoephedrine as indexes.Results:among components in Waigan Fenghan medicated bath formula,praeruptorin A,praeruptorin B,ephedrine,Pseudoephedrine and forsythin had obvious IL-6 regulatory activities;however,only ephedrine and pseudoephedrine had obvious peak in HPLC determination,their contents were 0.4215-0.4947 mg/mL and 0.1217-0.1915 mg/mL,respectively.Conclusion:Ephedrine and pseudoephedrine can be used as active compounds to evaluate Waigan Fenghan medicated bath formula’s IL-6 regulatory effect.
