EB病毒感染患儿T淋巴细胞miR-155、STAT1的表达及意义 被引量:5

Expression and significance of miR-155 and STAT1 in T-lymphocytes of children with Epstein-Barr virus infection

摘要 目的探讨Epstein-Barr病毒(Epstein-Barr virus,EBV)感染患儿CD4^(+)、CD8^(+)T淋巴细胞中微小RNA-155(MicroRNA-155,miR-155)、信号转导与转录激活因子1(Signal transducer and activator of transcription 1,STAT1)表达及其与EBV脱氧核糖核酸(EBV-deoxyribo nucleic acid,EBV-DNA)载量关系。方法选取在洛阳市妇幼保健院儿科治疗的85例急性EBV感染患儿为EBV感染组,及同期90例健康体检儿童为对照组。收集外周血单个核细胞(peripheral blood mononuclear cell,PBMC),采用流式细胞仪检测PBMC中CD3^(+)、CD4^(+)及CD8^(+)T淋巴细胞水平,计算CD4^(+)/CD8^(+)值。PBMC用免疫磁珠法分选得到CD4^(+)、CD8^(+)T淋巴细胞,采用实时荧光定量PCR检测CD4^(+)、CD8^(+)T淋巴细胞中miR-155、STAT1 mRNA表达及PBMC中EBV-DNA载量。相关性分析用Pearson检验。结果EBV感染患儿PBMC中CD3^(+)、CD8^(+)T淋巴细胞水平均高于对照组,CD4^(+)T淋巴细胞水平及CD4^(+)/CD8^(+)值均低于对照组(P<0.05)。EBV感染患儿CD4^(+)、CD8^(+)T淋巴细胞中miR-155、STAT1 mRNA表达均高于对照组(P<0.05)。EBV感染患儿CD4^(+)、CD8^(+)T淋巴细胞中miR-155、STAT1 mRNA表达水平均与EBV-DNA载量呈正相关(P<0.05)。结论EBV感染患儿CD4^(+)、CD8^(+)T淋巴细胞中miR-155、STAT1 mRNA表达均升高,二者高表达可能与EBV感染程度有关。 Objective To investigate the expression of microRNA-155(miR-155)and signal transducer and activator of transcription 1(STAT1)in CD4^(+)and CD8^(+)T-lymphocytes of children with Epstein-Barr virus(EBV)infection,and to analyze the correlation of the expression levels with EBV-deoxyribo nucleic acid(EBV-DNA)load.Methods Eighty-five children with acute EBV infection,who were treated in the Department of Pediatrics of Luoyang Maternal and Childre Health Hospital,were selected as the EBV infection group,and 90 healthy children in the same period were selected as the control group.Peripheral blood mononuclear cells(PBMC)were collected.The levels of CD3^(+),CD4^(+)and CD8^(+)T-lymphocytes in PBMC were detected by flow cytometry,and the CD4^(+)/CD8^(+)value was calculated.The CD4^(+)and CD8^(+)T-lymphocytes were isolated from PBMC by immunomagnetic beads.The expressions of miR-155 and STAT1 mRNA in CD4^(+)and CD8^(+)T-lymphocytes,as well as EBV-DNA load in PBMC was detected by quantitative real-time PCR.Pearson test was used for correlation analysis.Results The levels of CD3^(+)and CD8^(+)T-lymphocytes in PBMC in EBV infection group were higher than those in control group,and the level of CD4^(+)T-lymphocytes and CD4^(+)/CD8^(+)ratio in EBV infection group were lower than those in control group(P<0.05).The expression levels of miR-155 and STAT1 mRNA in CD4^(+)and CD8^(+)T-lymphocytes in EBV infection group were higher than those in control group(P<0.05)and the expression levels of miR-155 and STAT1 mRNA in CD4^(+)and CD8^(+)T lymphocytes of children with EBV infection were positively correlated with EBV-DNA load(P<0.05).Conclusions The expression levels of miR-155 and STAT1 mRNA in CD4^(+)and CD8^(+)T-lymphocytes elevated among children with EBV infection,and the high expressions may be related to the degree of EBV infection.
